G四联体稳定的脂质体DNA胶束用于细胞多色成像

【摘要】 脂质体键连的DNA链在水溶液中通过亲疏水作用自组装形成DNA胶束,DNA分子的可编程性及胶束的自组装特性使得DNA胶束在疾病诊断治疗中具有极大的潜力。然而,脂质体DNA胶束易于与血浆蛋白作用而裂解,易铆钉在细胞膜上而难于进入细胞,使得脂质体DNA胶束更广泛的应用受到限制。本文在脂质体与DNA链之间合成G碱基,胶束形成后,脂质体DNA链之间生成G四联体,G四联体的形成大大地提高了该胶束在血浆白蛋白溶液中的稳定性及跨膜穿透细胞的能力。通过混合组装带有可荧光共振能量转移染料的脂质体DNA分子,实现了细胞内的多色成像。
【Abstract】 Because of small size(<50nm),ability to self-assembled and programmable of DNA,lipid-based DNA micelle provides an option for diagnostic and therapeutic applications.However,their use has been limited by the fact of fast disruption by binding with albumin in serum and anchoring on the surface of cell membrane.In this paper,lipid-based DNA micelles were engineered by using of short G-quadruplex forming oligonucleotide extensions.Stability results of these particles show excellent ability of enough G base number(≥4) in avoiding disruption to serum protein and cell membrane.Finally,this system was used in intracellular multiplexed imaging.
