聚甲基丙烯酰胺衍生物类键合型高效液相色谱手性固定相的制备与手性识别性能研究

【摘要】 许多带有手性侧链的聚甲基丙烯酰胺可作为高效液相色谱的手性固定相用于极性手性化合物的拆分,特别是手性药物是拆分[1]。对手性化合物进行有效拆分的关键是发展高效的手性固定相[2]。本研究首先制备了含有少量三乙氧基硅侧链的聚甲基丙烯酰胺共聚物,然后通过三乙氧基硅侧链的分子间缩聚反应将此共聚物键合到硅胶表面,制备成键合型手性固定相(见图1)。采用高效液相色谱,以含有氯仿和四氢呋喃的混合溶剂为流动相,评价了该键合型手性固定相的手性识别能力(见图2)。
【Abstract】 Many polymethacrylamides bearing optically active side chains have been synthesized and used as chiral stationary phases(CSPs) for high-performance liquid chromatography(HPLC). The key to chiral separation is the development of effective CSPs. In this study, optically active copolymers of N-[(R)-(+)-α-methylbenzyl] methacrylamide((R)-MPMA) and a small amount of N-(3-(triethoxysilyl)propyl) methacrylamide(3-TPMA) were prepared. Then the copolymers were immobilized onto silica gel via the intermolecular polycondensation of the triethoxysilyl groups.The chiral recognition ability of the immobilized-type CSPs, which could be used with the eluents containing chloroform and tetrahydrofuran, was evaluated by HPLC.
