多通道电化学免疫传感用于甲胎蛋白检测

【摘要】 本文构建了一种多通道电化学免疫传感用于肝癌肿瘤标记物甲胎蛋白的定量检测。此传感体系利用多通道丝网印刷电极,在工作电极固定上抗甲胎蛋白鼠单克隆抗体,之后用于捕获抗原甲胎蛋白,再利用抗甲胎蛋白的兔多抗与之特异性结合形成夹心免疫复合物,最后利用辣根过氧化物酶聚合体偶联的羊抗兔二抗与三明治夹心免疫复合物结合,使电流信号放大。丝网印刷电极和多通道电化学检测仪均为自主研制,能够满足多通道电化学检测。通过条件优化,该体系能够检测到甲胎蛋白的动态范围为64pg/m L-250ng/m L,最低检测线为56pg/m L。该传感检测体系具有灵敏度高,特异性强,操作简单以及仪器便携等优点。
【Abstract】 We have developed a multi-channel electrochemical immunosensing system for the detection of α-fetoprotein. We used a carbon screen-printed electrode(SPCE) arrays.Capture antibodies were immobilized on electrode arrays, and the AFP antigens and detection antibodies were conjugated, then the anti-rabbit IgG conjugated to poly-horseradish peroxidase combinate with the formed sandwich immunocomplexes were used for obtaining the current signal by the electrochemical immunoassay, which provides a sensitive, rapid and simple method. The concentrations of AFP could be assayed in the ranges of 250 pg.mL-1~ 1000 ng.m L-1. The dection limit is as low as 56 pg.m L-1.
