云南省区收集琥珀蚕的mtDNA COⅠ基因克隆和系统进化分析

【摘要】 从云南省文山地区收集到一种琥珀蚕,其成虫的形态特征与印度琥珀蚕(Antheraea assama)和台湾红目大蚕(Antheraea formosana Sonan)较为一致。利用DNA条形码技术对云南琥珀蚕线粒体细胞色素C氧化酶亚基I(mt DNA COⅠ)基因长约617 bp的片段进行克隆和测序(Gen Bank登录号:KJ146686)。基于mt DNA COⅠ基因序列并应用Kimura-2-Parameter模型计算云南琥珀蚕与印度琥珀蚕的遗传距离为0.007,与台湾红目大蚕的遗传距离为0.014,而与其他大蚕蛾科泌丝昆虫的遗传距离则在0.076~0.161之间。构建的NJ树中云南琥珀蚕与印度琥珀蚕也最先聚在一起,再与红目大蚕聚在一起,从分子水平证实三者属于同种。初步认为,云南琥珀蚕与印度琥珀蚕是同种,台湾红目大蚕是与之亲缘关系较近的新种或亚种。
【Abstract】 The adult forms among Antheraea assama collected in Wenshan Prefecture Yunnan province, India Antheraea assama and Taiwan Antheraea formosana are very similar. Three of them were identified basing on DNA barcoding technology. A 617 bp segment of the mitochondrial gene, cytochrome c oxidase Ⅰ(COⅠ), of this species was sequenced to create a reference sequence for DNA barcoding(Gen Bank accession number: KJ146686), which was comparison difference with the other two. The phylogenetic analysis was done with the homologous series of other Saturniidae insects. The genetic distance between Antheraea assama of Yunnan and Antheraea assama of India COⅠ sequences was 0.007 based on Kimura-2-parameter made, and with Antheraea formosana was 0.014, whereas with the range from 0.076 to 0.161 compared with other silk-producing insects of Satumiidae. In phylogenetic trees constructed using NJ algorithms showed that Yunnan Antheraea assama were closely related with India Antheraea assama first, then with Antheraea formosana. It confirmed three of them are still the same species at the molecular level.We suggest that Antheraea assama collected in Yunnan and India Antheraea assama are the same species, Antheraea formosana could be considered as a new species that was close
