糖芯片的研究进展及其在海洋糖药物研发中的应用潜力

【摘要】 糖芯片是一种高通量筛选糖分子与其配体结合作用的生物芯片技术。目前其已广泛应用于糖组学、新药研发、疾病诊断等诸多研究领域。海洋多糖由于其特殊的化学结构,表现出抗肿瘤、抗病毒、免疫调节、降血脂、抗凝血等多种药理活性。如何将糖芯片技术应用到海洋多糖药物研发中具有重要意义。本文从糖芯片的构建与检测两个方面对其研究现状进行综述,并通过糖与靶点蛋白相关作用层面阐述了该方法在海洋糖药物研发中的应用潜力。

【Abstract】 Glycan microarray technology has emerged as a high-throughput tool applied to screening the interactions between carbohydrates and their ligands.Currently,it has been widely used in the fields of glycomics,drug discovery,and diagnostics.Marine polysaccharides with specific chemical structures possess versatile pharmacological activities including antineoplastic,antivirus,immunoregulatory,anticoagulant,and antihyperlipemic activity,etc.In this case,it is quite imperative to apply this technology to the research and development of new marine polysaccharide-based drugs.This article focuses on the recent advances in construction and detection processes of glycan microarrays.Meanwhile,we demonstrate the potential applications of glycan microarrays for development of marine polysaccharide-based drugs in terms of the significant roles of this technology in pharmacological research and discovery of new drug targets.
