虚寒性出血症大鼠尿液代谢组学异常及姜炭干预作用的研究

【摘要】 目的:运用代谢组学的方法研究虚寒性出血症大鼠尿液的代谢表型变化及姜炭的治疗作用。方法:利用核磁共振氢谱(1H-NMR)技术建立大鼠尿液的代谢指纹谱。应用主成分分析(PCA)和偏最小二乘判别分析(PLSDA)研究正常对照组与模型组、模型组与姜炭/干姜给药组之间的代谢物谱差异,并通过变量重要性投影(VIP)在尿液中发现潜在的生物标志物,通过矫正率筛选出姜炭温经止血的药效生物标志物。结果:姜炭可以使虚寒性出血证大鼠发生紊乱的内源性生物标志物回归正常水平,筛选出与虚寒性出血症相关的7种生物标志物,其中乙酰乙酸、丙酮酸、氮氧三甲胺、牛磺酸、马尿酸、色氨酸为姜炭温经止血的药效生物标志物。结论:代谢组学研究能较全面地反应生物体的生理及代谢状态,为中医"证"的本质研究及药物药效评价提供了一种新方法。
【Abstract】 Objective:To investigate the urine metabolic profile of the deficiency-cold and bleed mouse.And the intervention effects of ginger Carbon.Methods:Proton nuclear magnetic resonance(1H-NMR)technique was used to analyze the rat metabonome in urine.Partial least squares discriminant analysis(PLS-DA)was processed to analyze the metabonome difference between the control and mode group.Seven potential biomarkers were selected,via the parameter of variable importance in the projection(VIP).Principal components analysis(PCA)was employed to process the data from the ginger carbon treatment group and the hemostatic effects of ginger carbon were investigated through the content of potential biomarkers.Results:Ginger charcoal can make the disorder endogenous biomarkers of the model mouse to normal levels.And acetoacetic acid,pyruvic acid,nitrogen oxygen trimethylamine,taurine,hippuric acid and tryptophan were the efficacy of hemostasis biomarkers.Conclusion:The results obtained suggested that metabolomics studies could better reflect the whole status of metabolism in bio-systems,and could be treated as a potential powerful approach in pharmacological studies and investigation of the essence of’syndrome’ in traditional Chinese medicine.
