香远合剂对桥本甲状腺炎模型大鼠Tpo-Ab、Tg-Ab的影响(摘要)

【摘要】 目的:本论文通过观察香远合剂对实验性自身免疫性甲状腺炎(EAT)大鼠的实验室指标以及对临床桥本氏甲状腺炎(HT)患者的诊疗和随访进行探讨分析,对香远合剂的组方的理论依据以及香远合剂对HT的作用机制进行探讨,为此方治疗HT提供理论依据。方法:动物实验,购买雌性SD大鼠60只,体重约130±10g,进行适应性饲养1周后随机分为4组:正常组15只;模型组15只;西药组15只;中药组15只。正常组给予普通饲料喂养,剩余3组造模:将甲状腺球蛋白100mg完全溶解于双蒸水溶液50ml中,取10ml与等体积完全弗化剂充分混合成油包水状,初次免疫,取0.5ml在颈、背、腿、腹腔等部位施以多点皮下注射,第2周起加强免疫,取甲状腺球蛋白10ml与不完全弗化剂10ml充分混合后施以多点皮下注射,以后每周一次,至第5周,期间配合高碘水(1L水中加入0.64g碘化钠配制)饮水喂养。第6周开始,正常组给予普通饲料及饮水饲养至9周;西药组给予雷公藤多甙片,研磨,温开水配成悬浊液每天给药前大鼠称重,根据体重给药。等量灌胃至10周:香远合剂组每日给予香远合剂液4ml灌胃至10周;模型组继续造模至10周。第11周将各组实验大鼠眼球取血后离心1500转获取血清,采用ELISA法检测TG-Ab,TPO-Ab。采用SPSS17.0统计软件数据分析,数据满足正态性和方差齐性,组间采用方差分析,所列数据采用均数±标准差(±s)表示。结果:香远合剂和雷公藤多苷片都可降低血清TPO-Ab、TG-Ab抗体水平,且香远合剂治疗效果好于雷公藤多苷片。结论:香远合剂可以降低EAT大鼠的血清甲状腺球蛋白抗体(TG-Ab)和甲状腺过氧化酶抗体(TPO-Ab)水平,以达到治疗目的。
【Abstract】 Objective:This article by observing the experimentation indexes of Xiangyuan mixture on the rats with experimental autoimmune thyroiditis laboratory parameters(EAT),discussing and analysing the treatments and follow-up on the patients with clinical Hashimoto’s thyroiditis(HT),exploring the theoretical basis for the component of Xiangyuan mixture and the mechanism of Xiangyuan mixture to HT,provides a theoretical basis for the treatment of HT.Methods:Animal experiments,buy 60 female SD rats,weighing about 130 ± 10 g,feed adaptively1 week,and randomly divided into four groups:normal group 15;model group 15;western medicine group 15;medicine group 15 only.Normal group is given normal diet,and modeling the remaining three groups:the 100 mg thyroglobulin completely are dissolved in 50 ml double distilled water,take 10 ml of which with an equal volume of complete Vladimir agent are mixed fully like oil-water,primary immunization,the neck,back,legs,abdomen and other parts are subject to multi-point subcutaneous injection with the above 0.5ml mixture,from the second week boost immunization,take 10 ml thyroglobulin and 10 ml incomplete Vladimir agent after mixed with multi-point subcutaneous injection once a week and feed with high iodine water,up to the fifth week(1L water is added to 0.64 g of sodium iodide).Starting from the 6th week,the normal group is given with a normal diet and water,to nine weeks;western medicine group is given with grinding Tripterygium glycosides tablets together with warm water,every day weight the rats before injection,the volume of which based on the body weight.Fed with the same volume to the 10 th week;Xiangyuan mixture group is given with 4ml Xiangyuan mixture daily to the 10 th week;modeling group is continued to model up to the 10 th week.At the 11 th week,after take the blood of the eyes for the rats in each group,centrifuged at 1500 rpm to obtain serum,detect TG-Ab,TPO-Ab by ELISA.Using SPSS 17.0 statistical software to analyse data,the data satisfy normality and homogeneity of variance,among groups with analysis of variance,the data presented with mean ± standard deviation(x±s).Results:Xiangyuan mixture and Tripterygium glycosides tablets can low down serum TPO-Ab,TG-Ab antibody levels,and the treatment of Xiangyuan mixture is far better than Tripterygium glycosides.Conclusion:Xiangyuan mixture may low down serum thyroglobulin antibodies of EAT rats(TG-Ab)and thyroid peroxidase antibodies(TPO-Ab)level to achieve a therapeutic purpose.Xiangyuan mixtures can reduce the infiltration of thyroid cells for EAT rats,relieving their inflammatory response.
