优选特定比例下甘遂半夏汤加减甘遂甘草反药组合对腹水大鼠利水作用的影响(摘要)

【摘要】 目的:通过选择前期实验中含甘遂甘草不同比例配伍甘遂半夏汤对腹水大鼠影响实验研究中有效且未表现出明显毒性的含甘遂甘草特定比例的甘遂半夏汤,进行甘遂半夏汤中甘遂甘草反药组合加减应用对癌性腹水模型大鼠利水作用相关指标的影响,探讨药效或毒性反应是否与反药组合相关。方法:将Wistar大鼠按体重随机分为空白组、模型组、呋塞米阳性药组、甘遂半夏汤全方组(比例全方组)、甘遂半夏汤全方去醋甘遂组(比例全方去醋甘遂组)、甘遂半夏汤全方去炙甘草组(比例全方去炙甘草组),甘遂半夏汤全方去草遂组(比例全方去炙甘草醋甘遂组),除正常喂养外,每天上午除空白组、模型组外,其余各组灌胃给药,空白组和模型组灌胃蒸馏水,灌胃体积均为1ml/100g。给药5天后,收集大鼠24小时尿量,给药9天后,所有大鼠禁食不禁水12小时,取材前后称量大鼠体重,间接测量腹水量,放完腹水后腹主动脉取血,离心检测肾素、血管紧张素Ⅱ、醛固酮、抗利尿激素等指标。结果:与空白组比较,模型组和各给药组大鼠体重在给药第1、4、8天均显著降低(p<0.01),模型组和各给药组大鼠腹围/体重比值均较空白组升高(p<0.05或p<0.01),模型组和各给药组大鼠净体重均较均较空白组降低有统计学差异(p<0.01),模型组、阳性药组、比例全方去炙甘草组和比例全方去草遂组大鼠尿量均较空白组降低有统计学差异(p<0.05或p<0.01),模型组、比例全方去炙甘草组、比例全方去醋甘遂组和比例全方去草遂组大鼠醛固酮的含量较空白组升高有统计学差异(p<0.01),模型组和比例全方去醋甘遂组大鼠雪中抗利尿激素的含量均较空白组升高有统计学差异(p<0.01);与模型组比较,给药第4天各给药组大鼠体重均升高有统计学差异(p<0.05或p<0.01),给药第1天和给药第4天,比例全方组、比例全方去炙甘草组、比例全方去醋甘遂组和比例全方去草遂组大鼠腹围/体重比值均较模型组降低有统计学差异(p<0.05或p<0.01),比例全方组和比例全方去醋甘遂组大鼠尿量较模型组升高有统计学差异(p<0.05),阳性药组、比例全方组、比例全方去炙甘草组和比例全方去醋甘遂组大鼠腹水量较模型组显著降低(p<0.01),阳性药组和比例全方组大鼠血中抗利尿激素的含量均较模型组降低有统计学差异(p<0.05);与比例全方组比较,比例全方去炙甘草组大鼠血中醛固酮的含量升高有统计学差异(p<0.05),比例全方去醋甘遂组和比例全方去草遂组大鼠血中醛固酮的含量较比例全方组有升高趋势但无统计学差异,比例全方去醋甘遂组大鼠血中抗利尿激素的含量升高有统计学差异(p<0.05),比例全方去炙甘草组和比例全方去草遂组大鼠血中抗利尿激素的含量较比例全方组有升高趋势但无统计学差异。结论:各给药组在改善腹水模型大鼠利水作用相关指标方面均未表现出明显的毒性,且比例全方组在改善利水相关指标方面的作用优于比例全方去掉其中一味反药或两味反药组。
【Abstract】 Objective:we choose the certain proportion of Gansuibanxia decoction which is valid to the malignant ascites rats and does not show significant toxicity in early experiments,and observe the effect of Gansuibanxia decoction plus-minus Gansuigancao anti- drug combination on diuretic effect in malignant ascites rats to explore whether the efficacy or toxicity associated with the anti-drug combination.Methods:The Wistar rats were randomly divided into blank group,model group,furosemide group,Gansuibanxia soup group,Gansuibanxia soup removed Zhigancao group,Gansuibanxia soup removed Cugansui group,Gansuibanxia soup removed Zhigancao and Cugansui group.In addition to normal feeding,every morning except for the blank group and model group,the rest of the group was given drugs,the control group and the model group was given distilled water,the volume is 1ml/100 g.Administered five days,all rats were fasted but except water for 24 hours to collect urine.Administered nine days all rats were fasted but except water for 12 hours,we need to weigh weight of rats.When we remove the ascites,we also need to weigh weight of rats.We use the weight before removing ascites minus weight after removing ascites to indirectly measure the amount of ascites.When we remove the ascites,we need to abdominal aortic blood,centrifuge testing renin,angiotensin Ⅱ,aldosterone,antidiuretic hormone and other indicators.Results:Firstly,compared with the blank group,model group and other administration group show significant decrease in body weight on the lst,4th,8th days(p <0.01),show increase in the ratio of abdominal circumference and body weight(p < 0.05 or p <0.01),and show decrease in net weight(p < 0.01).What’s more,model group,furosemide group,Gansuibanxia soup removed Zhigancao group,Gansuibanxia soup removed Zhigancao and Cugansui group show decrease in urine(p < 0.05 or p < 0.01);model group,Gansuibanxia soup removed Zhigancao group,Gansuibanxia soup removed Cugansui group,Gansuibanxia soup removed Zhigancao and Cugansui group show increase in content of aldosterone(p< 0.01);model group and Gansuibanxia soup removed Cugansui group show increase in blood levels of aldosterone(p < 0.01).Secondly,compared with the model group,all administration group show increase in body weight on the 4th days(p<0.05 or p< 0.01);Gansuibanxia soup group,Gansuibanxia soup removed Zhigancao group,Gansuibanxia soup removed Cugansui group,Gansuibanxia soup removed Zhigancao and Cugansui group show decrease in ratio of abdominal circumference and body weight on the 1st,4th days(p<0.05 or p<0.01);Gansuibanxia soup group and Gansuibanxia soup removed Cugansui group show increase in urine(p<0.05);furosemide group,Gansuibanxia soup group,Gansuibanxia soup removed Zhigancao group,Gansuibanxia soup removed Cugansui group show decrease in ascites(p<0.01);furosemide group and Gansuibanxia soup group show decrease in blood levels of aldosterone(p<0.05).Thirdly,compared with the Gansuibanxia soup group,Gansuibanxia soup removed Zhigancao group show increase blood levels of aldosterone(p<0.05);Gansuibanxia soup removed Cugansui group and Gansuibanxia soup removed Zhigancao and Cugansui group show increase blood levels of aldosterone but the difference is not significant;Gansuibanxia soup removed Cugansui group show increase in blood levels of antidiuretic hormone(p<0.05);Gansuibanxia soup removed Zhigancao group and Gansuibanxia soup removed Zhigancao and Cugansui group show increase in blood levels of antidiuretic hormone but the difference is not significant.Conclusion:Each treatment group does not exhibit significant toxicity on related indicators of improving diuretic effect in malignant ascites rats,and Gansuibanxia soup group shows better performance in these indicators than the group that removed one or two Anti-drugs.
