阿萨伊油中β-谷甾醇及总甾醇含量测定和抗氧化活性研究(摘要)

【摘要】 建立阿萨伊油中甾醇类成分的含量测定方法,并对阿萨伊油的抗氧化活性进行评价。采用高效液相色谱法建立了阿萨伊油中β-谷甾醇、总甾醇(β-谷甾醇、豆甾醇之和,以β-谷甾醇计)的含量测定方法:采用Purosper STAR LP C18(4.6mm×250mm,5μm)色谱柱,以乙腈-异丙醇(30:70)为流动相,流速1 mL·min-1,柱温为室温,蒸发光散射检测器漂移管温度70℃、雾化器流速2.0 L/min。。结果表明β-谷甾醇在0.7666.38μg范围内线性关系良好,回归方程为Y=1.639X+0.7027(r=0.9992)。平均加样回收率为95.7%(RSD 2.76%)。阿萨伊油中β-谷甾醇的质量分数为1.85 mg·g-1,总甾醇含量的质量分数为1.93 mg·g-1。同时采用总氧自由基清除能力(TOSC)法对阿萨伊油的抗氧化活性进行评价,结果表明样品具有显著的抗氧化能力,样品浓度和抗氧化能力具有较好的剂量一效应关系。

【Abstract】 To establish a method for the determination of the sterols of the oil in the freeze-dried acai(Euterpe oleracea Mart.) and evaluate its antioxidant activities.A saponification/extraction procedure and high performance liquid chromatography(HPLC) analysis method were developed and validated for the analysis of phytosterols in PEE(Petroleum ether extract).Separation was achieved on a Purosper STAR LP C18 column with a binary,gradient solvent system of acetonitrile,and isopropanol(v/v).Evaporative light scattering detection(ELSD)was used to quantify p-sitosterol and the total sterols.Peak identification was verified by retention times and spikes with external standards.Standard curves were constructed(R>0.999) to allow for sample quantification.Recovery of the saponification and extraction was demonstrated via analysis of spiked samples.The highest content of total sterols is p-sitosterol.The antioxidant activities of the extracts were evaluated using the total oxyradical scavenging capacity assay(TOSC assay).The result showed that the PEE exhibited significant antioxidant properties,sample concentration and the antioxidant capacity had a certain relevance.
