火木层孔菌桑黄醇提物HPLC指纹图谱研究

【摘要】 目的:建立10批不同产地火木层孔菌桑黄醇提物(EEPI)的高效液相指纹图谱,并对其质量进行评价。方法:采用SHISEIDO CAPCELL PAK C18色谱柱(4.6mmI.D.×250mm,5μm),以0.2%磷酸水-甲醇进行梯度洗脱,流速1.0mL/min,检测波长为395nm。以Inoscavin A为参比峰建立EEPI特征吸收峰共有模式,并通过峰重叠率计算、相似度评价系统、聚类分析等对其相似度进行评价。结果:在选定的色谱条件下,得到10批不同产地火木层孔菌桑黄指纹图谱,标定了9个共有峰,并建立了火木层孔菌桑黄的指纹图谱共有模式,相似度在0.652-0.995,聚类分析结果将10火木层孔菌桑黄分为三大类。结论:此方法简单、准确、重复性好,峰的分离度较好,为有效地评价火木层孔菌桑黄的综合质量提供了参考依据。

【Abstract】 Objective:To establish the high performance liquid chromatography(HPLC) fingerprint of ethanol extracts of Phellinus igniarius(EEPI),and to evaluate its quality.Methods:Adopted 0.2%phosphoric acid solution- methanol as mobile phase with a SHISEIDO CAPCELL PAK C18 column(4.6mmI.D.x 250 mm,5um),the flow rate was l.OmL/min,and the wavelength was 395 nm.Taking Inoscavin A as reference peak to establish common mode of characteristic absorption peak of EEPI,and evaluating its similarity by peaks overlap rate calculation,similarity evaluation system and cluster analysis.Results:Under the chromatographic conditions,HPLC fingerprint of EEPI was established,and nine common peaks were recognized.Established common mode of characteristic absorption peak of EEPI and its similarity was between 0.652 and 0.995.According to cluster analysis,10 batches of Phellinus igniarius were divided into three groups.Conclusion:The method is simple,accurate,good repeatability and separation,providing basis for the comprehensive quality evaluation of Phellinus igniarius.
