淫羊藿合女贞子对激素干预哮喘大鼠GR亚型及转录因子的影响

【摘要】 目的:研究哮喘大鼠模型地塞米松治疗不同阶段合用中药淫羊藿、女贞子对糖皮质激素受体亚型及转录因子的影响。方法:卵蛋白致敏、激发建立大鼠哮喘模型,腹腔注射地塞米松模拟激素干预过程,分别在激素干预不同阶段(撤停前、撤停中、撤停后)合用中药淫羊藿、女贞子,激素撤停后凋处死动物,检测血清GR亚型含量、肺组织GCR亚型mRNA表达及肺组织54种转录因子蛋白表达。结果:淫羊藿女贞子合激素干预Ⅰ组可显著抑制激素干预后血清GRα量显著降低,淫羊藿女贞子合激素干预3个组均可显著抑制激素干预后血清GRβ含量的显著增高。激素干预后肺组织GRα及GRβmRNA表达均显著抑制,淫羊藿女贞子合激素干预Ⅱ组可显著上调GRαmRNA表达,淫羊藿女贞子合激素干预3个组均可显著上调GRβmRNA表达。激素干预组肺组织有43种转录因子的蛋白表达较哮喘模型组显著下调,仅有1种转录因子的表达上调;与激素干预组比较,淫羊藿女贞子合激素干预Ⅰ、Ⅱ、Ⅲ组分别有49种、45种、46种转录因子的表达显著上调。结论:淫羊藿女贞子合用地塞米松的协同抗炎机制及抑制激素不良反应的作用与对GR亚型的影响及上调肺组织转录因子的表达有关。
【Abstract】 Objective:To observe the effects of the compatibility of Epimedium and ligustrum lucidum on subtypes of glucocorticoid receptor(GR) and transcriptional factor(TF) of asthmatic rats with glucocorticoid(GC) intervention.Methods:Dexamethasone was injected into asthmatic model in rats with the oral gavage of Epimedium and Ligustrum lucidum at different treatment stage of GC withdraw.The following testing indexes,including the content of serum Hpotype of GR and the expression of GR mRNA and TF protein of lung tissue,were observed.Results:Serum GRa of group I of YN and GC was significantly decreased than that of group GC,while GRP of group Ⅰ,Ⅱ and Ⅲ of YN and GC were significantly higher.The mRNA expressions of GRα and GRβ in lung tissue were significantly inhibited after GC invention.Group Ⅱ of YN and GC can significantly increase GRa mRNA expression,while group Ⅰ,Ⅱ and Ⅲ of YN and GC can significantly increase GRβ mRNA expression.Of a total of 54 TFs screened,44 were identified with differential activities,of which 43 showed increased activities,only 1 showed decreased activities,compared with group asthma.Group Ⅰ,Ⅱ and Ⅲ of YN and GC had 49,45 and 46 kinds of TF expression significantly increased respectively,compared with group asthma.Conclusion:The mechanism of synergistic antiinflammatory and inhibiting adverse effects of Epimedium and Ligustrum combined dexamethasone is related to GR subtypes and increasing the expression of TFs.
