针刺眼针穴区与非穴区对D-IBS模型大鼠结肠SP及NK1表达的影响

【摘要】 目的:探讨眼针穴区的特异性及眼针疗法对D-IBS的作用机制。方法:SPF级雄性Wistar大鼠48只,采用慢性应激结合束缚法建立D-IBS大鼠模型,随机分为正常组、模型组、穴区组和非穴区组。后3组先建立D-IBS模型,然后穴区组针刺下焦区、大肠区、肝区和脾区;非穴区组选取同名眼针穴区外3mm处针刺。测定各组大鼠结肠运动功能;采用免疫组化法检测各组大鼠结肠组织SP及其受体NK1的蛋白表达水平变化。结果:与正常组比较,模型组大鼠结肠运动功能明显加强(P<0.01),结肠组织SP及其受体NK1的表达水平均显著升高(P<0.01);和模型组比较,穴区组结肠运动功能明显降低(P<0.01),非穴区组无显著差异(P>0.05);穴区组结肠组织SP及其受体NK1的表达水平均显著降低(P<0.01),非穴区组无显著差异(P>0.05)。结论:针刺眼针穴区能够有效治疗D-IBS,其可能的作用机制之一与下调结肠组织中SP和NK1表达有关。而针刺眼周非穴区部位则无明显疗效,从而说明眼针穴区部位具有相对特异性。

【Abstract】 Objective:To explore the point specificity of eye-acupuncture and the mechanism of eye-acupuncture on diarrhea-predominant irritable bowel syndrome.Methods:Forty eight male Wistar rats of SPF grade were randomly divided into a normal group,a model group,eye-acupuncture point(AA) group and a non-point(NA) group,The D-IBS rat model was established with the combination methods of the chronic stress and binding limbs.The AA group was treated by acupuncture at low energizer area,large intestine area,liver area,and spleen area,and the NA group by acupuncture at 3mm apart from the same points are a mentioned above and normal group and model group with no intervention.Protein expressions of SP and NK1 in colon were detected by SABC immunohistochemistry method.Results:Compared with normal group,the SP and NK1 protein in colon mucosa,myenteric nerve plexus and hypo-mucosa nerve plexus increased significantly in model group,Compared with model group,SP and NK1 protein in colon mucosa decreased significantly in AA group.Compared with AA group,the SP and NK1 protein in colon mucosa increased significantly in the NA group.Conclusion:Eye-acupuncture has a good therapeutic effect on D-IBS.It is suggested that one of the mechanism is relate to restrain SP and NK1 in colon tissue.Non-point area of eyeacupuncture has no obviously therapeutic effect and so to illustrate the point specificity of eye-acupuncture.
