大菟丝子不同炮制品特征图谱及有机酸含量研究

【摘要】 目的建立大菟丝子生品和炮制品的特征图谱,探讨大菟丝子炮制前后化学成分的变化规律,采用HPLC法对其中的新绿原酸、绿原酸、隐绿原酸、咖啡酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸7个有机酸类成分进行定量分析比较。方法采用Odyssil C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈-0.05%磷酸溶液,梯度洗脱,检测波长325nm,流速1mL·min-1,柱温35℃;用国家药典委员会"中药色谱指纹图谱相似度评价系统2004A"处理分析。结果指标性成分在测定范围内与峰面积线性关系良好(r≥0.9970);不同来源的大菟丝子生品或盐炙品间的特征图谱相似度较高,但炮制前后指标性成分含量差异显著,并出现炮制后新绿原酸、隐绿原酸、3,4-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸呈倍数增长,而绿原酸、咖啡酸、3,5-二咖啡酰奎宁酸呈倍数下降的趋势,且咖啡酸含量下降最明显。结论采用特征图谱结合指标性成分含量测定的方法可以较好的控制大菟丝子药材及饮片的质量。
【Abstract】 Objective To develop the HPLC specific chromatograms of Cuscuta japonica before and after salt-processed,toexplore the regularity of component changes in processing of Cuscuta japonica,and use HPLC to quantitatively analyze and compare neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,coffeic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid,4,5-dicaffeoylquinic acid.Method The separation was performed on an Odyssil C18column(250mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.05%phosphoric acid in gradient elution,at the flow rate of 1.0 mL·min-1,the column temperature was set at 35℃ and the detection wavelength was at325 nm.To analysis the data with Similarity evaluation system for chromatographic fingerprintof traditionalChinesemedicine(2004A).Result s The linear correlation of major components are good(r≥ 0.9970).HPLCspeciflc chromatograms of Cuscuta japonica from different areas were ofhigh similarity,but the differences were obvious in contents of major components,the contens of neochlorogenic acid,cryptochlorogenic acid,3,4-dicaffeoylquinic acid,4,5-dicaffeoylquinic acid were increasing exponentially after processing,on the contrary were chlorogenic acid,coffeic acid,and 3,5-dicaffeoylquinic acid,and the component of coffeic acid changed obviously.Conclusion TheHPLC specific chromatogram and contents ofseven organic acidscan be used for the quality control of Cuscuta japonica.
