【摘要】 目的分离蝉蜕胃蛋白酶解物中抗凝活性组分。方法活化部分凝血活酶时间试验(APTT)及紫外280nm检测为指标,通过Sephadex G-50、G-25、G10三种葡聚糖凝胶分离蝉蜕胃蛋白酶解物中抗凝活性组分,考察该组分PT、APTT、TT、FIB四种体外抗凝血药效,并通过MALDI-TOF-MS质谱图分析其分子量。结果通过Sephadex G-50、G-25、G10三种葡聚糖凝胶分离得到的蝉蜕胃蛋白酶解物活性组分F2-2-2,与空白组比较,具有良好PT、APTT、TT、FIB药效,其分子质量为800-1500道尔顿。结论蝉蜕胃蛋白酶解物活性组分F2-2-2具有良好体外抗凝活性,具有后续研究价值。更多还原

【Abstract】 Objective:To purify the anticoagulant constituent from periostracum cicada pepsin hydrolysate.Methods:Take PT、 APTT、 TT、 FIB as the in vitro anticoagulant indexes and UV280 nm as polypeptidedetection method,use Sephadex G-50、 G-25、 G10 to purify anticoagulation constituent from periostracum cicada pepsin hydrolysate.And analyze the molecularweight by MALDI-TOF-MS.Result:After purification,the Anticoagulation constituent,F2-2-2,of which molecularweight ranges from 800 to 1500 D,is proved to have a wonderful in vitro anticoagulant activity through the four indexes mentioned above.Conclusion:the purified anticoagulation constituent of periostracum cicada pepsin hydrolysateare valuable to be the follow-up study objects in physicochemicalproperty and stability area.更多还原
