牛蒡苷元对H89诱导的SH-SY5Y细胞损伤的保护作用

【摘要】 目的观察牛蒡苷元对H89诱导的SH-SY5Y细胞损伤的保护作用。方法用PKA抑制剂H89处理SH-SY5Y细胞,建立受损伤的细胞模型。实验设置空白、模型、治疗、阳性对照4组,分别给与相应处理。应用免疫细胞化技术检测细胞中β淀粉样肽、神经营养因子-3的表达,用细胞凋亡染色技术观察细胞凋亡的情况。结果与模型组相比,治疗组细胞的β淀粉样肽的含量明显下降(P<0.05),神经营养因子-3的含量明显上升(P<0.01),凋亡细胞数量减少(P<0.01)。结论牛蒡苷元对H89诱导的SH-SY5Y细胞损伤有保护作用。
【Abstract】 Objective To investigate the protective effect of arctigenin against the SH-SY5 Y cell damage induced by H89.Methods The PKA inhibitor(H89) was added to SH-SY5 Ycells to generate the model of injured cells.In this experiment there were four groups,including:control group,model group,group with arctigenin therapy and Salvianolic acid B intervention group,Immunocytochemical staining was performed to determine the expression of beta-Amyloid(Aβ)and neurotrophin-3(NT-3) in SH-SY5 Y cells.Apoptosis assay of Hoechst33258 staining was used to observe neuronal apoptosis.Results Arctigenin decreased the expression of Aβ(P<0.05) and increased the expression of NT-3(P<0.01) in SH-SY5 Ycells compared with the control group.Arctigenin also inhibited apoptosis in SH-SY5 Y cells-exposed to H89(P<0.01).Conclusions Arctigenin exerts the neuroprotective effects on H89-induced SH-SY5 Ycell damage.
