海藻玉壶汤中海藻甘草反药组合加减应用的急性毒性实验研究

【摘要】 目的:比较海藻玉壶汤(生甘草)组、海藻玉壶汤(炙甘草)组、去海藻(生甘草)组、去海藻(炙甘草)组、去甘草组、去海藻甘草组不同组别的小鼠急性毒性,评价海藻甘草反药组合在复方中应用的毒性大小。方法:采用灌胃给药的方法,给与不同配伍组合不同剂量的海藻玉壶汤,计算出LD50值,不符合LD50评价条件的组别,采用连续给药28天,即累计半数致死量的方法来进行毒性大小的评价;眼球采血,通过检测小鼠血清中肝肾功能相关指标,评价其对小鼠肝肾系统的影响。结果:各给药组均未做出LD50,海藻玉壶汤(生甘草)组LD50(16)值为71.08g·kg-1,海藻玉壶汤(炙甘草)组LD50(3)值为68.28g·kg-1,去甘草组LD50(16)值为55.78g·kg,去海藻(生甘草)组LD50(22)值为72.27g·kg-1,去海藻甘草组LD50(21)值为79.62 g·kg,去海藻(炙甘草)组小鼠连续给药28天仍未达到半数致死量,最大耐受量为80g生药/ml;给药各组对小鼠的肝肾脏功能均未见明显影响。结论:给药各组LD50((?))数值均属无毒的范畴,且对小鼠的肝肾脏系统未见明显毒性影响。

【Abstract】 object:To compare the different groups of acute toxicity of the Haizao Yuhu decoction(Radix Glycyrrhizae),the Haizao Yuhu decoction(the prepared Radix Glycyrrhizae),the Haizao Yuhu decoction removed Sargassum(Radix Glycyrrhizae),the Haizao Yuhu decoction removed Sargassum(the prepared Radix Glycyrrhizae),the Haizao Yuhu decoction removed Radix Glycyrrhizae and the Haizao Yuhu decoction removed Sargassum and Radix Glycyrrhizae in mice,and evaluate the toxicity.Method:The mice was given drug through stomach perfusion in different pairs and different doses of the Haizao Yuhu decoction,and calculate the LD50.If the value does not match the LD50 evaluation conditions,we will use the continuous administration of drugs for 28 days,that accumulated half of the lethal to evaluate the toxicity;Thread detect the related index of liver and kidney functions and evaluate the affect of that.Results:The LD(50)(n) of the Haizao Yuhu decoction(Radix Glycyrrhizae) is 71.08g·kg-1,the LD(50)(n) of the Haizao Yuhu decoction(the prepared Radix Glycyrrhizae) is 68.28g·kg-1,the LD(50)(n) of the Haizao Yuhu decoction removed Radix Glycyrrhizae is 55.78g·kg-1,the LD(50)(n) of the Haizao Yuhu decoction removed Sargassum(Radix Glycyrrhizae) is 72.27g·kg-1.the LD(50)(n) of the Haizao Yuhu decoction removed Sargassum and Radix Glycyrrhizae is 79.62 g·kg-1.Because of the Haizao Yuhu decoction removed Sargassum(the prepared Radix Glycyrrhizae) yet reached half lethal dose in the case of continuous administration for 28 days,the maximum tolerance dose of that group is 80 g crude drug/kg;Each experimental groups were not showed a significant impact to the liver and kidney functions in mice.Conclusion:the LD(50)(n) of each experimental group belongs to the category of non-toxic,and not showed a significant impact to the liver and kidney system in mice.
