【摘要】 目的:通过甘遂半夏汤中甘遂甘草反药组合加减应用对癌性腹水模型大鼠肝肾功能的影响,探讨药效或毒性反应是否与反药组合相关。方法:将Wistar大鼠按体重随机分为空白组、模型组、呋塞米阳性药组、甘遂半夏汤全方组、甘遂半夏汤去醋甘遂组、甘遂半夏汤去炙甘草组,甘遂半夏汤去炙甘草醋甘遂组,除正常喂养外,每天上午除空白组、模型组外,其余各组灌胃给药,空白组和模型组灌胃蒸馏水,灌胃体积均为1ml/100g。给药9天后,所有大鼠禁食不禁水12小时,取血,离心检测肝肾功能相关指标。结果:与模型组比较各给药组血中AST、ALT不具有统计学差异(p>0.05)而AST/ALT比值仅甘遂半夏汤全方组较模型组降低且具有统计学差异(p<0.01),与甘遂半夏汤全方组比较甘遂半夏汤全方去炙甘草组、甘遂半夏汤全方去醋甘遂组和甘遂半夏汤全方去草遂组AST/ALT比值升高且具有统计学差异(p<0.05或p<0.01);与模型组比较,各给药组中仅甘遂半夏汤全方组血清中ALP较模型组升高且具有统计学差异(p<0.05),而甘遂半夏汤全方去炙甘草组、去醋甘遂组较甘遂半夏汤全方组低具有统计学差异(p<0.05);甘遂半夏汤全方组血中总蛋白的含量较空白组、模型组和甘遂半夏汤全方去草遂组升高且具有统计学差异(p>0.05),而血中ALB含量与A/G比值相一致,即与模型组比较甘遂半夏汤全方组升高且具有统计学差异(p<0.05或p<0.01),甘遂半夏汤全方去炙甘草组、甘遂半夏汤全方去醋甘遂组和甘遂半夏汤全方去草遂组均较甘遂半夏汤全方组降低且具有统计学差异(p<0.05或p<0.01);与空白组比较,模型组、甘遂半夏汤全方去醋甘遂组、甘遂半夏汤全方去草遂组血中BUN的含量升高且具有统计学差异(p<0.05或p<0.01),而与模型组比较,阳性药呋塞米组和甘遂半夏汤全方组血中BUN的含量降低且具有统计学差异(p<0.05),其它给药组与模型组比较不具有统计学差异;与模型组比较阳性药呋塞米组、甘遂半夏汤全方组血中CRE的含量升高且具有统计学差异(p<0.05),但与空白组比较无统计学差异。结论:甘遂半夏汤全方组和甘遂半夏汤全方去掉其中一味反药或两味反药组在肝肾功能方面均未显示出明显毒性,且含甘遂甘草反药组合的甘遂半夏汤全方对于保护肝肾功能方面的药效优于全方去掉其中的一味反药或两味反药,反药组合在全方中有可能有增强药效的作用。
【Abstract】 Objective By observing the effect of euphorbia pinellia decoction plus-minus euphorbia licorice anti- drug combination on hepatic and renal functions in malignant ascites rats to explore whether the efficacy or toxicity associated with the anti-drug combination.Methods The wistar rats were randomly divided into a blank group,model group、furosemide group、euphorbia and pinellia soup group,euphorbia and pinellia soup removed Zhigancao group、euphorbia and pinellia soup removed Cugansui group、euphorbia and pinellia soup removed Zhigancao and Cugansui group.In addition to normal feeding,every morning except for the blank group and model group,the rest of the group was given drugs,the control group and the model group was given distilled water,the volume is 1ml/100 g.Administered nine days,all rats were fasted but except water for 12 hours,take bleeding,centrifugation,detect relevant indicators of liver and kidney function.Results Compared with model group,the serum AST,ALT of the treatment group does not have a statistically significant difference(p> 0.05),the AST / ALT ratio only the group of euphorbia and pinellia soup decreased and statistically significant(p <0.01) compared with the model group.Compared with euphorbia and pinellia soup group,the AST/ALT ratio of euphorbia and pinellia soup removed Zhigancao group、euphorbia and pinellia soup removed Cugansui group、euphorbia and pinellia soup removed Zhigancao and Cugansui group increased and statistically significant(p <0.05 or p <0.01).Compared with model group,only serum ALP of euphorbia and pinellia soup group increased statistically significant(p <0.05),while serum ALP of euphorbia and pinellia soup removed Zhigancao group and euphorbia and pinellia soup removed Cugansui group decreased and statistically significant(p <0.05).The serum total protein of uphorbia and pinellia soup group increased statistically significant(p> 0.05) compared with the group of the blank、the model and the euphorbia and pinellia soup removed Zhigancao and Cugansui.The ALB blood content and A / G ratio is consistent that compared with model group euphorbia and pinellia soup removed Zhigancao group、euphorbia and pinellia soup removed Cugansui group、euphorbia and pinellia soup removed Zhigancao and Cugansui group decreased and statistically significant(p <0.05 or p <0.01) compared with the euphorbia and pinellia soup group.Compared with the control group,the serum BUN level of the model group、euphorbia and pinellia soup removed Cugansui group、euphorbia and pinellia soup removed Zhigancao and Cugansui group elevated has a statistically significant difference(p<0.05 or p <0.01),compared with the model group,the serum BUN level of positive drug furosemide group and euphorbia and pinellia soup group decreased and statistically significant(p <0.05),the other treatment group compared with the model group does not have a statistically significant difference.Compared with the model group,the level of serum CRE of positive drug furosemide group and euphorbia and pinellia soup group elevated and statistically significant(p <0.05),but no significant difference between the control group.Conclusion Euphorbia and pinellia soup group and whole decoction remove one or two anti-drug did not show obvious toxicity in the liver and kidney function,and euphorbia and pinellia soup that contains anti-drug combination of gansui and gancao has better protective efficacy of liver and kidney function than the whole decoction remove one or two anti-drug,anti-drug combinations in the whole prescription may have enhanced the role of efficacy.
