人参皂苷Rg3对大鼠奥沙利铂神经毒性的保护作用

【摘要】 目的:评价人参皂苷Rg3对大鼠奥沙利铂神经毒性的保护作用,并初步探讨其作用机制。方法:30只1 0周龄雄性SD大鼠,分成3组,奥沙利铂模型组、人参皂苷Rg3组、正常对照组,奥沙利铂模型组每周腹腔注射4mg/kg奥沙利铂2次,人参皂苷Rg3组除了同模型组一样腹腔注射奥沙利铂外,每周腹腔注射人参皂苷Rg3 10mg/kg二次,正常对照组每周腹腔注射同等量5%葡萄糖注射液,干预4周后,观察至8周。分别于4周、8周各组各处死5只大鼠,ELISA法检测血清神经生长因子(NGF),免疫组化法检测NGF表达,TUNEL法检测脊髓背根神经节(DRG)神经元细胞凋亡情况。结果:与正常对照组比较,模型组大鼠血清中NGF浓度明显降低(P<0.05),人参皂苷Rg3组NGF浓度较模型组显著回升(P<0.05)。与正常对照组相比,模型组平均NGF免疫组化表达光密度值明显降低(P<0.05),而人参皂苷Rg3组较模型组明显回升(P<0.05)。与正常对照组比较,奥沙利铂模型组造模4周、8周后,DRG神经元细胞凋亡率均明显增加(P<0.05);与模型组比较,人参皂苷Rg3组细胞凋亡减少(P<0.05)。结论:人参皂苷Rg3对大鼠奥沙利铂神经毒性具有一定的保护作用,可增加NGF的表达,减少DRG神经元细胞的凋亡。

【Abstract】 Objective:To explore the protective effect of ginsenosides Rg3 on experimental oxaliplatin-induced peripheral neurotoxicity,and to understand the preliminary mechanism.Methods:To copy SD rats mo4 el of oxaliplatin neurotoxicity by intraperitoneal injection twice weekly with 4mg/kg oxaliplatin.The group of ginsenosides Rg3 treated with ginsenosides Rg3 twice weekly with 10mg/kg in addition.The normal control group treated with the same volume 5%glucose.After treating 4 weeks,finished the observation to the eighth week.And then detected serum nerve growth factor(NGF) by ELISA,NGF expression in dorsal root nerve ganglion tissues by immunohistochemistry,dorsal root nerve ganglion(DRG) tissue neuronal apoptosis by TUNEL assay.Results:The level of serum NGF in model group is significantly lower than the control group,and higher in the protection group than the model group.The NGF expression in DRG tissue in the model group is lower than the control group sighiflcantly,and is raised in the protection group.Positive apoptotic area is increased in the model group than the control group,and is decreased in protection group.Conclusions:Ginsenosides Rg3 might protect the oxaliplatin-induced peripheral nerve injury by inhibiting the degeneration and apoptosis of DRG cells.
