A multiple KH domain protein is required for asexual development and infection-related morphogenesis in the rice blast fungus

【摘要】 KH domain-containing proteins are widely conserved and play diverse roles in post-transcriptional gene control.However,few studies deal with their roles in pathogenesis of plant pathogenic fungi.Here,we identified a novel virulence gene PCG6 that encodes a protein with five KH domains in the rice blast fungus Magnaporthe oryzae.Deletion of PCG6 led to severely attenuated virulence toward host plants.Microscopic observation showed that the deletion mutant was reduced in appressorium formation,penetration peg formation,and development of invasive hyphae.In addition,the PCG6 deletion mutant was also significantly reduced in vegetative hyphal growth and conidiation.Pcg6 encodes a cytoplasmic protein expressing in mycelia,conidia,appressoria,and infection hyphae.Moreover,Pcg6 was co-purified and physically interacted with several proteins involved in nonsense mediated mRNA decay(NMD).Interestingly,the expression level of GLD1,a gene for glutamate dehydrogenase,was markedly increased in the PCG6 deletion mutant as compared with the wild type.Consistently,over-expression of GLD1 in the wild-type strain exhibited similar phenotypes as the PCG6 deletion mutant.Taken together,we proposed that the multiple KH domain protein Pcg6 regulates the glutaminolysis by NMD to control infection-related morphogenesis and developments in the rice blast fungus.
