季铵化壳聚糖微凝胶的制备及其葡萄糖响应性释放行为研究

【Abstract】 In general,glucose-sensitive controlled delivery systems can be prepared with pH sensitive gels and glucose oxidase(GOD),since GOD can react with glucose to form gluconic acid which triggers the pH-sensitive swelling/deswelling of the gels in different glucose concentration.Chitosan is a kind of natural biodegradable polycationic polysaccharide,however,its poor pH responsibility limited the application in glucose sensitive drug release systems.In this study,to solve this problem,chitosan was chemically modified using cationic quaternary ammonium monomer(CQAM) and quatemized chitosan(HTCC) microgels were prepared.The results showed that CQAM was confirmed to be grafted onto the molecular chain of chitosan successfully by ~1H NMR analysis.With increasing degree of substitution,storage modulus(G’) and crosslinking density(X_c)of HTCC microgels decreased,but mesh size(L_c) increased.The drug release behavior of HTCC microgels with DS 197%exhibited obvious pH and glucose responsive release behavior.
