四氧化三铁基纳米药物载体用于四价铂前药传递及克服肿瘤耐药研究

【摘要】 铂类药物,含顺铂、卡铂和奥沙利铂占据临床抗癌药物用量之半壁江山。卵巢癌对顺铂比较敏感,然而其会随着药物的使用,产生耐药,这导致了化疗的失败。也因此,50年来,卵巢癌的死亡率一直没有变化。纳米技术为传输铂类药物至肿瘤部位,降低毒副作用和克服耐药提供了契机。我们在此报道利用四氧化三铁纳米粒子来传输四价铂前药克服卵巢癌的耐药性。细胞和动物水平的研究揭示了四氧化三铁纳米粒子具有优越的克服顺铂耐药能力。
【Abstract】 Platinum drugs including cisplatin, carboplatin and oxaliplatin have become the most extensively used anticancer drugs which occupy over 50% of all in clinc use. Ovarian cancer is sensitive to cisplatin treatment at the initial stage, while it develops drug resistance gradually, resulting in the failure of chemotherapy and the death rate of ovarian cancer has almost not changed for 50 years. Advanced nanotechnology provides the possibility of delivering platinum drugs directly to the tumor sites, reducing the side effects and overcoming resistance. Here, iron nanoparticles were developed to load platinum(IV) prodrugs for overcoming ovarian cancer resistance. A systematical evaluation of the in vitro and in vivo antitumor performance revealed that iron nanoparticles showed superior ability to overcome ovarian cancer resistance.
