用于磷酸化肽捕获和质谱检测的稀土亲和材料的合成与表征

【摘要】 针对蛋白质组学研究中磷酸化蛋白/多肽质谱分析检测所面临的巨大挑战(如磷酸化肽丰度低、化学计量比小、质谱不易识别和受非磷酸化肽、无机盐强干扰等),利用稀土离子对磷酸根部分的高选择性亲和作用和水解催化作用以及磁性氧化铁快速磁分离特点,设计合成了稀土基磁性亲和材料(如Fe3O4@SiO2@CeO2,Fe3O4@LnPO4(Ln=Eu,Tb,Er),γ-Fe2O3@REVO4(RE=Sm,Dy,Ho)等),表征了它们的形貌和表面结构特性,实现了对复杂生物蛋白样品酶解液中的磷酸化肽的高选择性捕获、快速磁分离和容易的磷酸化肽质谱标记。这些工作将有助于磷酸化蛋白质组学的研究。

【Abstract】 Aimed at the great challenges faced by MS analysis and detection of phosphoproteins and phosphopeptides in proteomics research, which deal with small quantity of phosphoproteins, the low stoichiometry of phosphorylation, the suppression effect by nonphosphorylated peptides and interferences of high levels of small molecules such as salts, surfactants and other contaminants, we designed and synthesized rare earth-based affinity materials such as Fe3O4@Si O2@Ce O2,Fe3O4@Ln PO4(Ln=Eu,Tb,Er),γ-Fe2O3@REVO4(RE=Sm, Dy, Ho) using high selectivity and hydrolytic catalysis of rare earth ions to PO3-4 group as well as fast magnetic separation of magnetic iron oxides. We characterized their morphologies and surface structure nature, and realized high selective capture, rapid magnetic isolation and easy MS identification of the phosphopeptides from a complicated enzymatic digest of biological protein samples. This work will contribute to the research of phosphorylation proteomics.
