【Abstract】 Cell surface glycans are involved in cell-cell interaction and communication, cell adhesion, and many developmental processes. More importantly, the abnormal alteration of glycan expression on cell surface is associated with plenty of diseases, especially in cancers. Therefore, sensitive detection of carbohydrates on living cells is of urgent need for basic science advancement and clinical diagnostics. In this paper, we report a novel lectin-based biosensor for competitive assay of glycan espression on living cells. The biosensor was prepared by using Nafion/Au nanorod(GNR) layer to capture concanavalin A(Con A). {QDs-BSA-Au-S-mannose} bioconjugate was also synthesized by employing the BSA-protected Au nanoparticles(Au NPs) as matrixes to load Quantum Dots(QDs) for fluorescence signal generation. Thiomannosyl dimer was further incubated for competitive binding electrode-modified Con A with cell surface mannose. This proposed biosensor provides a reliable approach for in situ evaluation of the glycan expression on living cells and may facilitate the medical diagnosis and treatment.
