【Abstract】 Aptame based rolling circle amplification(aptamer-RCA) was developed as a novel versatile electrochemical platform for ultrasensitive detection of cancer cells. The primer sequence on the nanoprobes, which were displaced by the target cells, mediated a RCA reaction that generated hundreds of copies of a circular DNA template. Detection of the Cd S nanoparticles modified on the signal DNA which was complementary to the copies of the circular DNA template allowed enormous sensitivity enhancement in the assay of target cells. Because of the wide availability of aptamers for numerous cancer cells, this method holds great promise in clinical application.
