基于ATP适配体片段重新结合的信号扩增电致化学发光生物传感器的设计

【Abstract】 A novel signal-on electrochemiluminescent(ECL) biosensor based on adenosine triphosphate(ATP)-induced recombination of its split aptamer chips was presented. A 27-base anti-ATP aptamer had been split into two chips: ss DNA1 and ss DNA2. The former had been immobilized onto the gold electrode surface via Au-thiol interaction and worked as the capture probe, whilst the latter one had been functionalized with ECL reagent(tris(2,2’-bipyridyl) ruthenium(II)-doped silica nanoparticles(Ru-Si NPs)) and worked as the detection probe. In the present of ATP, two chips will recombinate together to form an ATP aptamer and results in the immobilization of the Ru-Si NPs on the electrode surface. The ECL intensity from the electrode is then significantly enhanced. It is found that the ECL intensities had a linear relationship with the logarithm of ATP concentration from 1.0×10-12 to 5.0×10-10 mol/L. The sensitivity is sufficiently high for most practical samples suggesting that it is a promising and powerful method for molecules detection.
