氨基酸膨润土固定化褶皱假丝酵母脂肪酶的研究

【摘要】 碱性钙基膨润土是一种新型的阴离子型层状黏土材料,由于其特殊的阴离子交换性能,具有广阔的应用开发前景。为了进一步认识碱性钙基膨润土,拓宽其应用范围,本课题研究用氨基酸对其进行改性来制备氨基酸膨润土,并初步探索氨基酸膨润土为载体应用于脂肪酶的固定化。结果表明氨基酸膨润土是一种较好的固定化材料,酶与载体之间"柔性链"的长度对其酶活及重复使用性都有影响,以谷氨酸改性最好,用XRD及FTIR对谷氨酸膨润土固定化酶前后分别进行了表征,蛋白固载量及固定化酶的活力分别为6.3mg/g土和391.2U/g,重复使用六次可保持约68%的活力;非离子型表面活性剂可以激活酶,如曲拉通X-100和X-114,可使固定化酶活性提高约2倍,对固定化酶的最佳处理时间为1h。

【Abstract】 Alkaline Ca-bentonite is a new kind of layered clay minerals; it has great practical and bright market prospect beeause of its special anion exchange capability. For the purpose of knowing alkaline Ca-bentonite further and broadening its application, this study will discuss about the preparation of amino acid bentonite and preliminary exploration of its application in the immobilization of lipase. The results show that amino acid bentonite is a new kind of immobilization material, the " flexible chain " between lipase and the support has influence on the enzyme activity and repeated use, glutamic acid is most suitable while its protein loading and specific activity were 6.3mg/g and 391.2U/g support,which can maintain 68% activity after reusing six times. The immobilization of lipase onto the glutamic acid bentonite was confirmed by Fourier transform infrared(FT-IR) spectra and X-ray diffractometer. Moreever, nonionic surfactant can activate the enzyme, such as X-100 and X-114, the immobilized enzyme activity increases about 2 times while its best incubation time is 1 h.
