穿心莲内酯抗血管生成作用研究

【摘要】 目的:研究穿心莲内酯(Andrographolide,AD)对血管生成的抑制作用。方法:采用人脐静脉内皮细胞HUVEC模型,观察AD对HUVEC细胞迁移和VEGF蛋白表达的影响,同时考察AD对血管生成相关信号通路活化的影响。结果:AD在浓度1.25-5.0μmol·L-1,显著抑制内皮细胞的迁移和三维管腔结构的形成,Western blot结果表明其作用机制与抑制VEGF/VEGFR2信号通路的激活,并抑制转录因子NF-κB活化有关。结论:AD具有显著的抗血管新生活性,提示此可能为AD发挥抗肿瘤作用的机制之一,具有良好的应用开发前景。
【Abstract】 OBJECTIVE:To investigate the inhibitory effects of Andrographolide(AD) on angiogenesis.METHODS:Human umbilical vein endothelial cells were employed to investigate the migration of HUVEC and the expression of VEGF,as well as the activation on signaling pathway associated with angiogenesis.RESULTS:AD(1.25-5.0μmol·L-1) significantly inhibited cell migration and the formation of tubular three-dimensional structure,of which the mechanism was associated with the inhibition on activation of VEGF/VEGFR2 signaling pathway and activation of transcription factors NF-κB,showed by the results of Western blot.CONCLUSION:AD showed obvious inhibition on angiogenesis,which may be one of the mechanisms of the anti-tumor effect of AD.AD was of good application and development prospects.
