皂角刺总黄酮提取物平衡溶解度及油水分配系数的测定

【摘要】 目的:测定皂角刺总黄酮提取物的平衡溶解度及表观油水分配系数,为剂型设计提供依据。方法:以槲皮素为指标,采用HPLC法测定皂角刺总黄酮提取物在水、0.1 mol.L-1HCl和不同p H值磷酸盐缓冲液中的浓度,采用摇瓶法测定其在正辛醇-水中的表观油水分配系数。结果:37℃下,以槲皮素为指标,皂角刺总黄酮提取物在水中的平衡溶解度为10.67 mg.L-1;表观油水分配系数为0.78(log P app=-0.11),磷酸盐缓冲液中,表观油水分配系数随p H值的增大而呈现先减小后增大的趋势。结论:皂角刺总黄酮提取物的脂溶性较好,其平衡溶解度和表观油水分配系数受p H的影响较大,为缓控释制剂研究提供基础。
【Abstract】 Objective:To determine the equilibrium solubility of Gleditsia sinensis extracts and its apparent oil/water partition coefficient in different solvents,so as to provide a basis for dosage from design.Methods:Quercetin was taken as the examination target,an HPLC method was performed to detect the equilibrium solubility in water,0.1 mol.L-1HCl and the medium of different p H(2-10) values and apparent partition coefficient for the n-octanol-water/buffer solution system.Results:The equilibrium solubility of quercetin was10.67 mg.L-1 in water at 37 ℃,the apparent oil/water partition coefficient was 0.78(log P app=-0.11).The apparent oil/water partition coefficient was firstly decreased and then increased with the increase of PH value in the phosphate buffer solution.Conclusion:The fat solubility of Gleditsia sinensis extracts was good,its equilibrium solubility and apparent oil/water partition coefficient were influenced greatly by the p H values.
