【摘要】 目的以瓜蒌薤白为基方,结合现代临床用药组成活血化痰方,研究该方水提和醇提两种工艺提取物对心梗大鼠心功能及心脏结构的影响,探讨《金匮要略》瓜蒌薤白类方佐伍白酒煎服的科学依据。方法活血化痰方分别进行水提醇沉、60%醇提工艺进行提取,提取物按人等效剂量饲喂心肌梗死大鼠,分别在心梗术后8日和22日两个时间点运用超声技术检测大鼠心功能;术后23天取心脏组织,进行HE和Masson染色,分析心肌组织病理变化。结果饲喂60%醇提取物组大鼠在心梗术后8日即出现阻止心梗后心脏EF、ES值下降趋势,但与模型组比较不具统计学意义(P<0.05),术后22日时本组大鼠EF、FS值下降程度明显降低,与模型组比较具有显著统计学意义(P<0.01);其作用与减轻心肌组织纤维化程度(P<0.05),减少心肌坏死面积(P<0.01)相关。水提醇沉物虽然能阻止EF下降,降低心肌梗死面积、减轻心肌纤维化程度的作用趋势,但与模型组比较差异无统计学意义(P>0.05)。结论由瓜蒌薤白为基础的活血化痰方60%醇提物较之水提取物具有明显干预心肌梗死后大鼠心脏恶性重塑,阻止心功能下降的作用,证明《金匮要略》中瓜蒌薤白类方伍白酒煎服的科学意义,为传统方剂的现代应用奠定基础。更多还原

【Abstract】 OBJECTIVE Huoxuehuatan prescription consisted of Combined Trichosanthes kirilowii Maxim with Allii macrostemi bulbus were used as the basic medicines to reatment the syndrome of phlegm and stagnated blood of coronary heart diseases.To study the influence of the aqueous extract and alcohol extract from Huoxuehuatan prescription on rat myocardial infarction cardiac function and structure,explore the scientific basis of Trichosanthes kirilowii Maxim with Allii macrostemi bulbus matching baijiu decoction in Synopsis of Golden Chamber.METHODS Huoxuehuatan formula is extracted by water and ethanol precipitation or 60% alcohol,feeding myocardial infarction rats with the equivalent dose extract.detecting the cardiac function in rats by using ultrasound at 8 and 22 d.Taking heart for HE and Masson staining and then analysising the pathological changes of myocardial tissue at 23 d.RESULTS The rats which feed with 60% alcohol extract appear to prevent EF,ES value decline after 8th,but there is no significant difference compared with model group(p<0.05).Rats EF,FS value decline significantly lowercompared with model group(p<0.01) at 22 d;It’s relevant to reducing myocardial tissue fibrosis(p<0.05)and infarct(p< 0.01).Water extract may prevent EF decline,reduce myocardial infarct area,and fibrosis,but there is no statistically significant difference compared with the model group(p>0.05).CONCLU SION Huoxuehuatan phlegm which is based with Trichosanthes kirilowii Maxim and Allii macrostemi bulbus that60% alcohol extract than water extract has obvious effect on ischemic myocardial remodeling and preventing cardiac function decline,It is proved that the scientific significance of Trichosanthes kirilowii Maxim with Allii macrostemi bulbus matching baijiu decoction in Synopsis of Golden Chamber and lay a foundation for the modern application of traditional formula.更多还原
