HPLC法同时测定癫痫患者卡马西平、奥卡西平及其主要代谢物的血药浓度

【摘要】 目的:建立高效液相法同时测定人血浆中卡马西平、奥卡西平及其主要代谢物10,11-环氧卡马西平、利卡西平浓度。方法:以盐酸普萘洛尔为内标,血浆经甲醇沉淀后直接进样。色谱柱采用岛津ODS C18柱(4.6*50mm,3um),流动相为乙腈:0.25M甲酸溶液(20:80)。结果:在线性范围内线性良好(R~2>0.9994),回归方程分别是Y=0.0795254X-0.00128006、Y=0.0255575X-0.00102140、Y=0.0519381X-0.00246967、Y=0.0239854X-0.000662765,各待测样品的日内、日间RSD 0.50%-16.77%,相对回收率90.7%-103.1%,绝对回收率90.0%-106.8%。结论:本方法操作简单,结果准确,适用于卡马西平和奥卡西平的日常血药浓度监测。
【Abstract】 Objective:To establish the HPLC method for the simultaneous determination of carbamazepine,oxcarbazepine,and their main metabolites(carbamazepine-10,ll-epoxide,monohydroxy derivative) in plasma samples.Methods:The analytes were extracted by protein precipitation,and propranolol hydrochloride was used as internal standard.Results:the linearity of our method checked in their plasma concentration ranges was excellent,and their calibration curves were Y=0.0795254X-0.00128006 、 Y=0.0519381X-0.00246967 、Y=0.0255575X-0.00102140 、 Y=0.0239854X-0.000662765,The RSDs of intra-and inter-day precision ranged from0.50%-16.77%,the extraction recovery ranged from 90.0%-106.8%and the relative recovery ranged from90.7%-103.1%.Conclusion:this method is proved to be convenient,accurate and suitable to measure concentrations of the analytes in clinical samples.
