两株多重耐药嗜芳香环弧菌的耐药分子分析

【摘要】 从患病的半滑舌鳎分离得到两株菌株JS291和JS295,采用生化实验和分子生物学方法(弧菌特异性基因16S r RNA、HSP60和mre B)鉴定为嗜芳香环弧菌,对其进行耐药性检测及耐药分子机制分析。通过K-B纸片扩散法和肉汤稀释法检测菌株对抗菌药物和消毒剂的耐药性,通过PCR扩增和测序法检测菌株中I、II、III类整合子并对其携带基因盒进行分析。结果表明,两株弧菌对环丙沙星、链霉素、复方新诺明、利福平、三氯异氰脲酸、苯扎溴铵等产生耐药。两菌株均携带I类整合子、qac EΔ1和sul1耐药基因;菌株JS291和JS295均携带基因盒,分别为dfr16-aad A2和arr-3-dfr A27,前者介导对甲氧苄啶和氨基糖苷类耐药,后者介导对利福平和甲氧苄啶耐药。这提示,两株菌株呈多重耐药性,并且多重耐药表型与整合子-基因盒密切相关。
【Abstract】 The drug resistance and the molecular mechanism of two Vibrio strains JS291 and JS295 were studied. The two strains isolated from diseased Cynoglossus semilaevis were identified as Vibrio cyclotrophicus by a biochemical test combined with a molecular method. The K-B disk diffusion method and the broth dilution method were used to determine the antimicrobial susceptibility of the strains to different antibiotics and disinfectants. The PCR amplification and DNA sequencing were used to detect class I, II, and III integrons and the gene cassettes. The results indicate that the two strains were resistant to ciprofloxacin, streptomycin, cotrimoxazole, rifampicin, trichloroisocyanuric acid and benzalkonium bromide. Class I intergron、qac EΔ1 and sul1 genes were present in both two strains. The integron-gene cassettes of JS291 and JS295 were encoding dihydrofolatereductase(dfr16), aminoglycoside-(3’’)(9)-adenylyltransferase(aad A2), rifampin ADP-ribosylating transferase(arr-3) and dihydrofolate reductase(dfr A27), respectively. These findings suggest that the two strains exhibited multi-drug resistance; the multi-drug resistance phenotype may be closely related to the integron-gene cassettes.
