利用RNA-Seq技术从大黄鱼性腺中发现性别差异基因
Sex-biased Gene Discovery from Gonadal Transcription Analysis by RNA-Seq in Large Yellow Croaker(Larimichthys crocea)

【摘要】 鱼类性别一直是科学家研究的重点,因为研究鱼类性别不仅具有很大的科研价值,而且具有重要的实践意义。大黄鱼主要分布于东部沿海地区,是中国海水网箱养殖量最大的重要经济物种,其生长具有雌雄二态性,雌性个体比雄性个体长得更快。但是目前关于大黄鱼性别分化的研究却很少。在该研究中,利用RNA-Seq技术分析正在发育中的大黄鱼雌雄性腺的基因表达谱。我们以大黄鱼综合转录组作为参考,将测序共产生的40G的读长映射到77405个转录本。基因差异分析发现4713个基因在XY性腺中特异表达,46个基因在XX性腺中特异表达。该研究探索了大黄鱼转录组水平上的基因表达谱,为大黄鱼早期性别分化时期关于功能分化和分子机制的研究提供了必要基础。
【Abstract】 Sex in fish has long been a hotspot topic in research communities. The investigation of sex in fish is of great scientific value, but also has important practical significance. The large yellow croaker(Larimichthys crocea) mainly distributes in coastal regions of East Asia and is one of the most economically important marine fish in China. The growth of the female large yellow croaker is much faster than those of male individuals. However, the gene expression differences of male and female gonads in the large yellow croaker were rarely reported. In this study, the gene expression profiles in male and female gonads of developing large yellow croaker individuals were analyzed by RNA-Seq technology. Taking the comprehensive transcriptome of the large yellow croaker as a reference, 77,405 transcripts were mapped by about 40 Gb reads. The gene differential expression analysis showed that 4,713 genes were specifically expressed in male gonads, while 46 genes were specific to female gonads. This study explored gene expression profiles in large yellow croaker gonads at the transcriptome level, providing essential foundation for the following gene function analysis and the molecular mechanism investigation of early-stage sex differentiation in large yellow croaker.
