重金属Cu胁迫对半叶马尾藻(Sargassum hemiphyllum)生长及生化组分的影响

【摘要】 本文采用半静态实验方法研究了半叶马尾藻(Sargassum hemiphyllum)对重金属铜(Cu)胁迫的生理响应。实验设置4个胁迫组,Cu浓度分别为0.01、0.05、0.25和1.25mg/L,不加Cu为对照组,在0d、2d、4d和8d时分别测定藻体鲜重、叶绿素a(Chla)和类胡萝卜素(Car)含量、超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性及丙二醛(MDA)含量。结果表明:随Cu浓度增大藻体相对生长速率(RGR)呈先升后降的趋势,其中以0.05mg/L胁迫组为最大;Cu≤0.05mg/L的胁迫组,Chla和Car含量均恢复到对照组水平(P>0.05),SOD活性相对稳定;而Cu>0.05mg/L的胁迫组,胁迫2d后,Chla和Car含量均低于对照组,1.25mg/L胁迫组的SOD活性显著高于对照组(P<0.05),至8d时,CAT活性和MDA含量均显著高于对照组(P<0.05),且1.25mg/L胁迫组的MDA含量显著下降(P<0.05)。因此,当Cu≤0.05mg/L时,在一定程度上可促进藻体生长,抗逆能力无明显变化;而Cu>0.05mg/L时,藻体生长和抗逆能力均降低。

【Abstract】 In order to study the growth and biochemical composition responses of Sargassum hemiphyllum to Cu stress, the thallis under four treatments with different Cu concentrations: 0.01, 0.05, 0.25 and 1.25 mg/L, together without Cu as the control, were incubated in laboratory with a semi-static test for 8 days. The fresh weight, chlorophyll a(Chla), carotenoid(Car), superoxide dismutase(SOD), catalase(CAT) and malondialdehyde(MDA) of thalli were determined in 0d, 2d, 4d and 8d. The results showed that: with the increasing Cu concentration, the RGR of thalli was increased until Cu concentration was 0.05mg/L, and then decreased. The content of Chla and Car, in Cu≤0.05mg/L groups, were recovered to the control level, SOD activity was relatively stable. However, after 2d, the content of Chla and Car were lower than the control in Cu>0.05mg/L groups, and SOD activity was higher than the control significantly when Cu concentration was 1.25mg/L(P<0.05); and to 8d, CAT activity and MDA content were higher than the control significantly(P<0.05), and the MDA content of 1.25mg/L was dropped significantly(P<0.05). Therefore, it was suggested that the growth of thalli was promoted to some extent and the resilience was no change significantly in Cu≤0.05mg/L; however, it was inhibited and the resilience was weakened in Cu>0.05mg/L.
