鲑鱼鱼精蛋白絮凝活性和抑菌活性研究

【摘要】 本文探究了鱼精蛋白对微藻、细菌和真菌的絮凝效果以及对12种细菌的抑菌效果。结果表明鱼精蛋白对盐藻的絮凝作用最好,20μg/m L的鱼精蛋白对盐藻的絮凝率可以达到85.51%。将絮凝后的盐藻沉淀细胞和上清液分离,并对残留在上清液中的盐藻细胞进行再培养和再絮凝,再次絮凝可以得到相似的絮凝效果(85.89%)。观察显微镜下的盐藻沉淀细胞,沉淀细胞可以保持较好的活性和游动状态。抑菌实验表明鱼精蛋白对革兰氏阳性菌和革兰氏阴性菌均具有较好的抗菌活性。通过CM Sepharose Fast Flow阳离子色谱柱对鱼精蛋白进行分离纯化,并对纯化后的鱼精蛋白进行絮凝和抑菌实验,表明峰IV具有较好的絮凝作用和抑菌作用,峰ⅡI只具有较弱的抑菌作用而没有絮凝作用。对峰ⅡI和峰IV进行氨基酸组分分析得到,峰ⅡI和峰IV分别富含赖氨酸和精氨酸。鱼精蛋白的絮凝活性和抑菌活性使其成为一种理想的絮凝剂和抑菌剂。
【Abstract】 The flocculation effect of salmine on microalgae, bacteria and fungus and the antibacterial effect of salmine on 12 kinds of bacteria were investigated in this study. Salmine showed the best flocculation effect on Dunaliella salina. The flocculating rate on D.salina could reach 85.51% at 20 μg/m L. The precipitated cells of D.salina and supernatant after flocculation were separated and the residual cells in the supernatant were re-cultured and re-flocculated. Re-cultured residual cells could be flocculated by salmine at the similar flocculating rate(85.89%). The precipitated cells were observed by the microscope, which could maintain good vitality and swimming state. Besides, salmine exhibited antimicrobial activity against gram positive and gram negative bacteria. After salmine being purified by CM Sepharose Fast Flow ion exchange chromatography, PEAK IV showed flocculating activity and antimicrobial activity, whereas PEAK ⅡI showed weak antimicrobial activity but no flocculating activity. Amino acid analysis showed that PEAK ⅡI and PEAK IV were rich in lysine and arginine, respectively. The flocculating and antimicrobial properties indicate that salmine can act as an ideal flocculant and bacteriostatic agent.
