红笛鲷TRAF3基因的克隆、表达及其功能研究

【摘要】 TRAF3(Tumor Necrosis Factor receptor-associated factor 3)是TRAF家族成员之一,能够与胞内多种受体如TNFR、NLR、TLR/IL-1R等相互作用,进而调节NF-κB和MAPK信号通路的活性。为研究TRAF3在鱼类免疫信号转导中的作用。本研究利用RACE技术克隆了红笛鲷TRAF3(Ls-TRAF3)基因全长,通过蛋白结构域分析发现Ls-TRAF3在N端含有1个环指结构,2个锌指结构,C端还有该家族保守的coiled coil和MATH结构域。荧光显微镜观察显示Ls-TRAF3为全细胞分布。Ls-TRAF3在所有组织中均有表达,尤其在脾、头肾和肝脏中表达量较高。在哈氏弧菌、poly I:C分别刺激后,Ls-TRAF3在脾脏中表达量均有不同程度的提高,说明Ls-TRAF3可能参与了红笛鲷的免疫应答过程。上述研究结果为进一步研究TRAF3的功能以及鱼类的免疫机制奠定了基础。
【Abstract】 TRAF3 is a member of TRAF family, which served as the signal transducer to mediate the activation NF-κB and MAPK by interacting with TNFR 、NLR、TLR /IL-1R. To study the function of TRAF3 in immune signal transduction, we cloned the red snapper(Lutjanus sanguineus) TRAF3(Ls-TRAF3) c DNA, Sequence analysis showed that Ls-TRAF3 contains four characteristic domain, including an N-terminal RING finger, two zinc finger, a coiled tail motif and a conserved MATH domain. Fluorescence microscope observation showed Ls-TRAF3 was distributed in whole cells. Ls-TRAF3 transcripts expresses ubiquitously in all tissues tested, with the higher expression in spleen、head kidney and liver. Furthermore, Ls-TRAF3 m RNA expression significantly increased in spleen after infection with Vibrio harveyi or poly I:C, which indicating that Ls-TRAF3 might be involved in the host’s defense against pathogen infection.These results provide some evidence for exploring the mechanism of fish immune response.
