利用SSR分子标记分析陆地棉亲本遗传距离与杂种优势的关系
Analysis on Correlation between Genetic Distance and Heterosis in Upland Cotton with SSR Molecular Marker

【摘要】 实验采用8个陆地棉材料按照NCⅡ设计配制15个F1,测定亲本及F1的油分和蛋白质含量及铃重、衣分和纤维品质,并分析了F1代的中亲和超亲优势。利用SSR分子标记,用118对多态性引物对亲本及其F1进行扩增,按照Nei和Li公式计算亲本间的遗传相似系数(GS)和遗传距离(GD),结果显示:8个亲本间的遗传相似系数变幅为0.612~0.992,平均值为0.682。8个亲本可分为3类。分子标记聚类结果与材料的遗传来源背景及系谱关系基本一致。亲本SSR遗传距离与F1断裂比强度的高亲优势呈显著负相关;与铃重和马克隆值的高亲优势都达到了显著的正相关,与纤维长度的中亲优势、高亲优势都达到了显著和极显著的正相关;与F1的衣分呈极显著的负相关,和衣分高亲优势呈极显著的正相关,与其中亲优势相关不显著。
【Abstract】 The content of cottonseed oil and protein,fiber quality and yield traits were investigated in 8 upland cotton parents and their 15 hybrids derived from the NCⅡ mating design.118 pairs of primers were amplified among 8 parents and their F1.Genetic similarities and genetic distance among the 8 parents were calculated which ranged from 0.612 to 0.992 with an average of 0.682.The cluster analysis showed that the 8 parents could be classified into 3 distinct clusters which correspond to their hereditary background and their pedigree analysis.The correlation between the genetic distance and heterobeltiosis of fiber strength showed significant negative but significant positive between the genetic distance and heterobeltiosis of boll weight and Micronaire value.There is positive and significant positive correlation among fiber strength mid-parent heterosis and heterobeltiosis and genetic distance.On lint percent,there is significant negative correlation between F1 value and genetic distance,significant positive correlation between the heterobeltiosis and genetic distance,uncorrelated with their mid-parent heterosis.
