贵州省2010-2012年间羊种布鲁氏菌分离株的遗传与分子流行病学特征研究

【摘要】 目的了解贵州省2010-2012年间羊种布鲁氏菌分离株的遗传与分子流行病学特征,为贵州省人间和动物间布病疫情的预防和控制提供科学依据。方法应用鲁氏菌属特异性PCR(BCSP31-PCR)和种/型特异性PCR(AMOS-PCR)对贵州省2010-2012年间分离的6株布鲁氏菌进行复核鉴定,采用MLVA-16方法分析经复核的菌株,并将其与来自我国其他省份的菌株进行聚类分析。结果来自贵州省患者和动物(羊)的6株布鲁氏菌株经BCSP31-PCR鉴定为布鲁氏菌属细菌,AMOS-PCR将其鉴定为羊种布鲁氏菌,MLVA-16分析显示6株菌株与布鲁氏菌各种型代表菌株中的羊种生物3型布鲁氏菌聚类最近,但在部分VNTR位点上存在重复数目差异,贵州省羊种布鲁氏菌与来自云南、福建、广东省羊3型布鲁氏菌遗传关系最近。结论贵州省2010-2012年间的布鲁氏菌分离株均为羊种生物3型布鲁氏菌,但菌株间存在遗传多样性,与来自云南、福建和广东省羊3型布鲁氏菌的遗传关系最近,该研究结果将为贵州省人间和动物间布病疫情的预防和控制提供科学依据。

【Abstract】 Objective To understand the genetic and epidemiologic characteristic of B.melitensis strains isolated in Guizhou Province within 2010-2012,and to provide scientific basis for the control and prevention of human and animal brucellosis in Guizhou Province.Method Brucella genus specific BCSP31-PCR and species-specific AMOS-PCR were used to identify the bacteria strain,and the identified strains were analyzed using MLVA-16 and cluster analysis of B.melitensis strains from Guizhou Province and other province were conducted.Result Six Brucella strains were identified as B.melitensis using BCSP31-PCR and AMOS-PCR.MLVA-16 analysis showed that the bacteria strains were most closely clustered with B.melitensis biovar 3,and differences of repeat number at part of VNTR loci were also displayed.Six strains were closely related to B.melitensis isolates from Yunnan,Fujian and Guangdong Province.Conclusion PCR methods combined MLVA-16 identified the six Brucella strain isolated in Guizhou Province in 2010-2012 as B.melitensis biovar 3,and the genetic diversity of the stains were also showed.Six strains were closely related to the isolates from Yunnan,Fujian and Guangdong Province.The results of this study provided scientific basis for the control and prevention of Brucellosis in Guizhou Province.
