二甲双胍对大鼠肾小球系膜细胞氧化应激的影响

【摘要】 目的:观察二甲双胍对高糖培养的大鼠肾小球系膜细胞(MCs)氧化应激的影响,探讨其保护肾脏机制。方法:大鼠肾小球系膜细胞分别培养在正常对照组(NC组)、高糖(HG组)及高糖+不同浓度二甲双胍组。48h后,比色法测定细胞上清液超氧化物歧化酶(SOD)、丙二醛(MDA)含量,RT-PCR方法测定细胞p22phox和p47phox mRNA表达情况。结果:与NC组比较,HG组MCs p22phox和p47phox mRNA表达明显增加,上清液SOD活力明显降低,MDA含量明显增高(P<0.05)。二甲双胍干预高糖组上述变化较HG组明显受抑制(P<0.05),且与浓度相关。结论:二甲双胍可降低大鼠肾小球系膜细胞高糖介导的NADPH氧化酶表达,减轻氧化应激,该作用可能与其保护肾脏部分有关。
【Abstract】 Objective To observe the effects of metformin on oxidative stress in rat glomerular mesangial cells(GMCs) cultured by high glucose and explore its reno- protective mechanisms.Methods GMCs were cultured in normal glucose concentration(group NC),high glucose concentration(group HG)and different concentrations of metformin(group M1,M2 and M3)plus high glucose.After 48 h later,the activities of the supernatants superoxide dismutase(SOD) and the malondialdehyde(MDA) were analyzed by using colorimetry.The expressions of p22 phox and p47 phox mRNA in GMCs were detected by reverse transcription- polymerase chain reaction.Results:(1) Compared to the group NC,the expressions of p22 phox and p47 phox mRNA significantly increased,the activities SOD decreased while the MDA level greatly increased in group HG(P <0.05);(2) Compared with group HG,the levels of intracellular p22 phox and p47 phox mRNA and the supernatant MDA level were decreased,while SOD activities increased significantly in group M1、M2 and group M3(P <0.05) with dose.Conlusions Metformin can inhibit their expressions of NADPH oxidase and reduce the levels of oxidative stress in rat GMCs,which may be related to partly its reno- protection.
