【摘要】 目的:探讨血管内皮生长因子(VEGF)936C/T突变与VEGF表达水平及其与早期动脉硬化(ATH)的关系。方法:参照国际超声目前常用诊断标准和中国高血压病防治指南,选取无亲缘关系的颈动脉内膜增厚患者184例(ATH组)和健康对照组人群240例(NC组),采用PCR-RFLP方法确定了全部病人基因型,对不同组间的936C/T多态性以及不同基因型组临床与生化指标以及VEGF浓度进行了统计分析。结果:ATH组CC基因型及C等位基因频率显著高于健康对照组,而ATH组(CT+TT)基因型频率及T等位基因频率显著低于NC组(x~2=18.956,P<0.01和x~2=18.417,P<0.01)。VEGF表达水平自TT基因型组至CT基因型组至CC基因型组逐步升高(P<0.01或P<0.05)。结论:中国汉族人群中存在VEGF936C/T突变,VEGF936C等位基因及CC基因型可能是中国汉族人易于发生早期动脉硬化的的危险性遗传标志,而VEGF936T等位基因和携带T等位基因的基因型(936TF基因型和936CT基因型)可能是与早期ATH发病风险降低相关。更多还原

【Abstract】 Objectives:To study the association of C936 T polymorphism at 3’- untranslated region of human vascular endothelial growth factor(VEGF)gene and atherosclerosis(ATH).Methods:The normal control group consisted of 120 subjects and 184 patients with thickened CIMT were recruited into this study.We refer to the carotid intima- media thickness(CIMT) above 0.9 mm was considered indicative of thickened CIMT according to current sonographic criteria.Genotypes were identified by PCR- RFLP among all the subjects,serum VEGF levels were tested using ELISA.Results:The frequencies of genotype CC and allele C were significantly higher in ATH group than in NC group,however the frequencies of genotype(TT + CT) and allele T were significantly lower in ATH group than that in NC group(χ~2= 18.956 and χ~2 = 18.417,P <0.01 and P< 0.01 respectively).CONCLUSIONS:Allele 936 C of VEGF may be a genetic marker susceptible to ATH.while allele 936 T may be a protective genetic marker of ATH,The 936C→T mutation of VEGF may be a protective factor against ATH.更多还原
