Ti-Si-N功能涂层制备及细胞相容性研究
Deposition and cell compatibility of Ti-Si-N coatings for biomedical application

【摘要】 心血管系统疾病目前主要依靠人工移植体来治疗。钛合金是设计制造人工移植体的理想材料,为了有效保护人体组织和延长人工移植体的使用寿命,一般均在钛合金基体上制备具有良好生物相容性的功能涂层。本文采用磁控溅射方法在Ti6Al4V钛合金表面制备了不同Si含量的Ti-Si-N涂层。用XRD,XPS和AFM分析手段对Ti-Si-N涂层进行了微观结构分析,结果发现随着Si含量的增加,氮化钛晶体的取向发生改变,同时涂层的表面粗糙度变小。对涂层的水接触角测量显示随着Si含量的增加,涂层的亲水性并没有大幅度的改变。而Ti-Si-N涂层的溶血率随着粗糙度的降低而降低,均不超过5%,表明Ti-Si-N涂层对血细胞的损伤率极低。在不同Si含量的Ti-Si-N涂层样品上培养血管内皮细胞6天后,细胞增殖良好,不同Si含量的Ti-Si-N涂层均不会抑制血管内皮细胞的生长,并且在涂层上紧密接触生长,具有良好的铺展度。因此,Ti-Si-N涂层未来有可能作为心血管系统人工移植体的理想生物功能涂层。
【Abstract】 Implantable and extracorporeal cardiovascular devices are commonly made from titanium alloy. In order to improve the biocompatibility and the service life of these devices, such as coating method has been applied to the surface of titanium alloy.Superhard ternary Ti- Si- N coatings with different Si content are deposited on titanium alloy substrates by magnetron sputtering and cell compatibility is studied. The microstructures of all the coatings were characterized by X- ray photoelectron spectroscopy(XPS), X- ray diffraction(XRD), and atomic force microscope(AFM). The crystal orientation of TiN is changed were found for Si content in the range 28-41 at%. Meanwhile, surface roughness(RMS) value of Ti- Si- N coatings is becoming smaller. In vitro studies show that the surface hydrophilicity and hemolysis are about the same as those of the titanium alloy control. Vascular endothelial cells cultures reveal better proliferation on the coatings compared to the titanium alloy control and the Ti- Si- N coating with 28 at% Si exhibits excellent cell spreading. In conclusion,Ti- Si- N coatings have large potential in cardiovascular applications.
