日本囊对虾野生与选育群体遗传多样性的微卫星分析
Microsatellite analysis on genetic diversity of wild and breeding stocks of Marsupeneaeus japonicus

【摘要】 利用微卫星分子标记技术对日本囊对虾霞浦野生群体(XP,34尾)和选育群体(G2,34尾)的遗传多样性进行分析。野生群体采自福建霞浦三沙海域,选育群体取自福建省东山县茂鑫水产有限公司养殖的G2日本囊对虾。5个微卫星位点在两个群体中共检测到88个等位基因,多态信息含量介于0.6542-0.9076之间,均为高度多态位点。野生群体与养殖选育群体平均等位基因数为16和8.4,平均有效等位基因数为15.4133和5.7086,平均杂合度为0.7353和0.5529,平均多态信息含量为0.8479和0.7230,群体间遗传分化指数Fst为0.0404。结果表明,日本囊对虾野生群体和养殖选育群体的遗传多样性都较为丰富,尚未发现明显的遗传分化现象。
【Abstract】 Five microsatellite markers were used to study the genetic diversity of the wild Marsupeneaeu japonicas from Xiapu coastal area in eastern Fujian and the breeding stock from Fujian Dongshan MAO XIN Fisheries Ltd. The genetic DNA was extracted from 34 individuals for wild stock and breeding one respectively. 88 alleles were obtained and the polymorphism information content(PIC) value per loci was greater than 0.5, indicating that these five loci were highly polymorphic. The average number of alleles of wild stock and breeding stock were 16 and 8.4, and the average effective number of alleles were 15.4133 and 5.7086. The mean heterozygosity were 0.7353 and 0.5529, while the mean polymorphism information content were 0.8479 and 0.7230.The coefficient of genetic differentiate(Fst) was 0.0404. The results showed that the genetic diversity of wild stock and clutured one were relatively abundant, there was no significant genetic differentiation existed.
