培养基成分对牡丹愈伤组织褐化的影响
The Effect of Medium Components on the Browning of Callus in Subculture of Tree Peony(Paeonia suffruticosa)

【摘要】 以牡丹品种‘珊瑚台’鳞芽诱导组培苗的叶柄愈伤组织为材料,探讨不同浓度激素组合、铁盐、无机盐、琼脂以及pH值等培养基成分对愈伤组织继代培养褐化的影响。结果表明:不同梯度NAA结合6-BA和KT,珊瑚台愈伤组织的褐化情况表现出不同变化规律,呈现对KT更为敏感,以NAA0.5mg·L-1+KT0.3mg·L-1处理中褐化率最低,为13.3%。在不同无机盐和铁盐组合中,以为1/4MS+1/4Fe的褐化率最低。pH为6.5时,培养30d后褐化率为0;以6.0g·L-1琼脂浓度下愈伤组织生长最好,培养30后的褐化率为5%。得出牡丹愈伤组织继代培养的最佳培养基配方为:1/4MS+1/4Fe+KT 0.3mg·L-1+NAA 0.5mg·L-1+PVP1g·L-1+LH 0.5g·L-1+蔗糖30g·L-1+琼脂6g·L-1,pH为6.5。
【Abstract】 The callus of Tree Peony(Paeonia suffruticosa) ’ Shan Hu Tai ’ that induced from the petiole of plantlets were used as materials in this research,to study the effects that the medium content of different concentration of hormone combination,iron salt,inorganic salt,Agar and PH on the browning of callus in subculture.Results showed that followed the combination of different concentration of NAA,6- BA and KT,the browning of the callus of Paeonia suffrulicosa ’ Shan Hu Tai ’ showed different change rule,and KT performed more sensitive,with the additives of NAA(0.5mg · L-1) + KT(0.3mg · L-1),the browning rate reduced to 13.3%.In different combination of inorganic salt and iron salt,the browning rate was the lowest at the combination of 1/4MS + 1/4Fe.Under the pH of 6.5,after the callus was culture for 30 d in the dark environment,the browning rate was 4%;As the concentration of Agar was 6.Og · L-1,the growth of the callus was the best,and the browning rate was only 5%after it was culture for 30 d.Concluded from the experiment,the best culture medium formula for the peony callus in subculture was:1/4MS + l/4Fe + KT0.3mg · L-1 + NAA0.5mg · L-1 + PVP1 g · L-1 + LH0.5g · L-1+ Sugar30 g · L-1 + Agar 6.0g · L-1 pH6.5.
