中原牡丹品种主要花香挥发物的多元统计分析

【摘要】 本研究比较15个中原牡丹品种鲜花花瓣花香挥发物的组分和含量,并进行主成分分析和聚类分析。用顶空-固相微萃取(HS-SPME)方法收集、提取花香挥发物并利用气相色谱质谱联用(GC-MS)技术分离与鉴定花香挥发物的成分及其含量,用SAS9.0统计软件对分析数据进行主成分分析和聚类分析。结果表明,共检测出42种物质成分,其中26种在牡丹花香挥发物中属首次报道。不同品种间各组分的含量差异较大。主成分分析表明,前5个主成分累计贡献率达81.34%,β-苯乙醇、月桂醇、正十二烷、γ-依兰油烯和对甲氧基苯分别在前5个主成分中的荷载值最高,可确定为对牡丹花香挥发物组分贡献最大的物质成分。总方差50%以上的贡献来自第1和第2主成分,可以认为正己醇、乙酸叶醇酯、乙酸己酯、香叶醇、香茅醇、丙酸香茅酯、1-正十三烷醇和Z-7-十四碳烯醇等醇和酯也是牡丹花瓣花香挥发物的重要成分。各品种的主成分得分值分析结果与感官评价结果相一致,‘景玉’‘小海黄’‘水晶白’‘豆绿’‘花二乔’和‘蓝宝石’的得分高于其他品种。研究表明,主成分得分的二维分布图对区分供试品种花香挥发物组成的相似性及差异性具有简便直观的特点,系统聚类在揭示供试品种相对遗传距离及亲本利用上有一定参考价值,两种不同方法的分析结果基本一致,认为牡丹花香挥发物的组分及含量可能与其颜色特征有关系。两种方法既有共性又有特异性,都能较好地为牡丹品种花香的改良和亲本利用提供科学依据。

【Abstract】 The floral volatiles and their concentrations in fresh petal of 15 different tree peony cultivars belonging to central plains of China were compared,and then were analysed by principal component analysis(PCA) method and cluster analysis(CA) method.The floral volatiles were collected and concentrated using head space solid-phase microextraction(HS-SPME)and then separated and identified using GC/MS,The dataset included analytical values from all volatile components measured.PCA and CA in SAS9.0 was used to evaluate the dataset The results showed that fourty-two volatiles were identified,of which26 were identified for the first time in tree peony blossoms.The volatile components and their contents of each cultivar differ greatly.PCA of blossom volatiles demonstrated that the first five PCs explained 81.34%of the cumulative.Benzeneethanol,duodecyl alcohol,dodecane,gamma-muurolen and p-dimethoxybenzene had the highest loading values,and they could be conformed to be the largest contributors to tree peony floral volatiles.As for the first two eigenvectors explained more than 50%of the total variance,hexanol,cis-3-hexenyl acetate,hexyl acetate,trans-geraniol,citronellol,citronellol propionate,tridecanol,Z-7-tetradecenol could also be the important compounds for tree peony floral volatiles.The PC scores of every sample corresponded to the results of organoleptic evaluation,that is ’Jingyu’,’Xiaohaihuang’,’ Shuijingbai’,’Doulv’,’Huaerqiao’ and’ Lanbaoshi’ had higher score than other cultivars.The similarities and differences among cultivars could easily be found using PCA score plot for each sample in the first two dimensions,and the relative genetic distance among samples and the use of parents could be evaluated using CA.Though they are different methods,the analysis results were agreed that tree peony blossom volatiles may relate to morphological characteristics,such as floral color.There were both general and specific character between the two methods,and they could provide scientific basis for the improvement of tree peony floral volatiles or parents using.
