细胞分裂素和蔗糖浓度对草莓花药培养愈伤组织起源影响的组织学观察

【摘要】 草莓花药培养组织起源是能否获得单倍体的关键。本试验通过对不同的激素和蔗糖浓度的花药培养过程的组织学观察,发现了在低细胞分裂素浓度(1.0mg/L)和蔗糖浓度(3%、6%)下,组织基本上都是起源于药隔和花丝断口处,而高的细胞分裂素浓度(3.0mg/L和5.0mg/L)和蔗糖浓度(9%、12%和15%)可以完全抑制花药壁和花丝断口处愈伤组织的产生,但对药隔愈伤组织的产生抑制不完全,在15%的蔗糖浓度下仍然有药隔细胞启动分化,但愈伤组织的增殖得到抑制。本试验虽然有花粉细胞启动分裂,然后分生组织化,但是没有观察到花粉组织产生。

【Abstract】 Callus origin of strawberry anther culture is the key for haploid induction.In this test,by observing the anther culture process in different hormone and sucrose concentration,callus were basically originated from the connective and filament fracture in the low 6-BA(1.0mg/L) and sucrose concentration(3%,6%) and were completely inhibited in filament fracture and anther wall in high 6-BA(3.0mg/L and 5.0mg/L) and sucrose concentration(3.0mg/L and 5.0mg/L),but were can incompletely inhibited in the connective,there were differentiated callus in anther wall in 15%sucrose.High sucrose concentrations can inhibit callus proliferation.Although pollen cells has been observed to start to split,and then meristem,but not observed to produce pollen callus.
