利用Pi9基因和MAS技术培育高抗稻瘟病水稻新品系

【摘要】 利用已克隆的广谱抗稻瘟病基因Pi9的DNA序列,设计功能分子标记pi9utr和Ins2-1,通过标记基因型辅助选择和连续回交育种实践,定向改良34份籼稻受体亲本的稻瘟病抗性。获得如下结果:pi9utr、Ins2-1是两个高效显性分子标记,在.Pi9供体亲本75-1-127与受体亲本间存在明显而稳定的多态;室内接种和田间病圃表型验证表明,两个分子标记的辅助选择效率均可达100%;基因表达分析表明,回交群体中所有抗病个体均能检测到Pi9基因的高效表达,所有感病个体均无Pi9基因的表达;通过连续多代回交结合标记辅助选择,分别获得了与11份受体亲本遗传背景非常相似的数十个高抗稻瘟病纯系,及其余23份受体亲本的不同世代回交群体,为进一步培育广谱高抗稻瘟病水稻新品种(组合)奠定了基础。

【Abstract】 To improve blast resistance of thirty-four receptor indica rice lines,two DNA markers,pi9 utr and Ins2-l,were developed based on the DNA sequence of the cloned broad-spectrum blast resistance gene Pi9,and marker-assisted selection breeding practice was implemented in this study.The main results were as follows:pi9utr and Ins2-1 were dominant markers,showing stable polymorphism between the Pi9 donor 75-1-127 and most receptors.Selection efficiency by two markers for blast resistance phenotype were 100%,by indoor inoculation and nursery phenotypic confirmation.The Pi9 gene showed high level expression in all resistant plants,according to randomly sampled individuals analyses of two backcross populations.Dozens of new rice lines with high-level blast resistance were developed from eleven receptors respectively,and more backcross populations with different generations by other twenty-three receptors were constructed,which paved a solid way for further breeding elite blast resistant rice varieties.
