表达葡萄糖氧化酶基因抗晚疫病马铃薯的培育

【摘要】 本文通过根癌农杆菌介导,用葡萄糖氧化酶(GO)基因转化马铃薯栽培品种大西洋(Atlantic)和夏普蒂(Shepody),获得了23个转基因植株。经卡那霉素的抗性筛选,PCR 扩增检测,植物总 DNA 斑点杂交和 Southern 杂交分析,表明 GO 基因已整合到转基因马铃薯的基因组中。通过转基因马铃薯离体叶片抗晚疫病菌(Phytophthora infestans)试验及植物体内葡萄糖氧化酶基因表达释放 H2O2的分析,结果表明转基因马铃薯植株的离体叶片比未转基因植株叶片的发病时间延迟而且发病程度降低,其抗病能力与 H2O2释放量呈正相关。

【Abstract】 Two commercial potato cultivars“Atlantic”and“Shepody”were trans- formed with glucose oxidase gene from Aspergillus niger via Agrobacterium tumefaciens mediated gene transfer.23 regenerated transgenic potato plants were obtained.Kanamycin resistance,PCR amplification,nucleic acid spot hybridization and southern blot analysis showed that the glucose oxidase gene was integrated into tranformed potato genome.South- ern blot analysis revealed that there were 2-4 copies of glucose oxidase gene in each potato tetraploid genome.Detached transgenic potato leaves were inoculated with spors of phyto- phora infestans.Tests showed that the number and the size of lesions was reduced and the appearance of lesions was delayed in some transgenc plant leaves.
