Expression of TRAIL and caspase 3 and their significance in rat’s renal ischemia-reperfusion injury

【摘要】 Objective: to observe the expression changes of TRAIL(TNF-related apoptosis-inducing ligand),caspase-3 and apoptotic express after rat’s renal ischemia and reperfusion, to investigate their roles in renal ischemical reperfusion injury. Methods: 48 SD maleness rats are separated random into sham group and ischemical reperfusion group, and execute every 6 rats after 6h.12h.24h.72h ischemical reperfusion. We observe the pathological changes of nephridial tissue with light microscope and immunohistochemistry, and to observe the expression of the TRAIL and caspase 3 with immunohistochemitry, and to detect the apoptosis of nephridial tissue with TUNEL. Results: the form and structure of nephridial tissue are normal, there are a sprinkle expression or without expression of TRAIL, caspase 3 and apoptosis in sham group. There are different level’s lamellar necrosis and inflammatory cell infiltrate in IR group. TRAIL, caspase 3 and apoptosis are significantly advanced than sham group(P < 0.01).There are changes of nephridial tissue microarchitecture in different period of acute ischemic ischemic and reperfusion with serious damage of renal function, and the expression of TRAIL and caspase 3 have corresponding changes. The immunohistochemistry shows that there are main expressions of TRAIL and caspase 3 in distal convoluted renal tubule and collectors ,which arrived peak after 24 hour, and there are a gleam of expressions in acinus renis without marked changes. With the expressions elevating of TRAIL and caspase 3, the apoptosis rates of every IR group appear homologous advancing. The expressions of apoptotic and TRAIL .caspase 3 demonstrate show prominent coincidence with TUNEL. Conclusion :In rats acute ischemical reperfusion injury, the high expression of TRAIL which cause the advance of caspase 3 is one of important path of the cell apoptosis after renal ischemia-reperfusion injury, and at same time, it may play important roles in early renal ischemia-reperfusion injury and its severity.
