用单细胞凝胶电泳法检测人造矿物纤维对DNA的损伤

【摘要】 目的比较研究10种人造矿物纤维(MMF)对人淋巴细胞的损伤作用。方法采用日本纤维状物质研究协会提供的10种标准人造矿物纤维(JFM),用单细胞凝胶电泳法,以人淋巴细胞为靶细胞,用迁移率和迁移度评价对细胞DNA链断裂程序。结果RF1、RF2、RF3、MG、SC1、TO1引起DNA链断裂程度与对照比较,统计学上有显著性差异。其中RF1、RF2、RF3和MG1纤维所引起DNA链断裂程度与温石棉接触。结论 10种人造矿物纤维均可引起不同程度的DNA链损伤,但损伤程度除耐火纤维外,均低于温石棉。

【Abstract】 Objective This study is to compare DNA injury induced by 10 kinds of man-made mineral fibers (MMMF)provided by Japan Fibrous Material Research Association (JFMRA)with UICC standard chrysotile in human lymphocytes.Methods Single-cell gel electrophoresis(SCGE) assay was used to detect DNA injury.Indexes of migration rate and migration degree were used to express the extent of DNA strand breaks.ResuIts The results showed that the DNA injury induced by refractory fibers (RF1,RF2,RF3)was approximate to that of chrysotile.The extent of DNA strand breaks induced by RF1, RF2 and micro glass fiber(MG1) was also similar to that of chrysotile.Other types of MMMF had lower genotoxicity.Conclusion All of 10 kinds MMMF could induce DNA strand breaks in different degrees,but the effect was lower than that of chrysotile fiber.
