【摘要】 目的:建立柱前衍生化高效液相色谱法测定人唾液中多胺含量的方法。方法:唾液经纯化、丹磺酰氯衍生化后用苯提取,以 Hypersil BDS CN 为正相色谱柱,丙酮—氯仿(2:98,v/v)为流动相, 检测波长为425nm,流速1 ml/min。结果:多胺中腐胺和精胺均在50～1000nmol/ml 范围内线性关系良好(r=0.9991;r=0.9992),方法回收率分别为94.65%和90.48%,RSD 为1.6%和2.1%.结论:方法灵敏、稳定,重现性好.检测人唾液中多胺含量具有临床实用意义。更多还原

【Abstract】 OBJECTIVE:To develop a high-performance liquid chromatographic method for the determination of polyamine in person saliva.METHODS:The derivatization with dansyl chloride was followed by normal—phase chromatography with a mobile phase of acetone/chloroform(2:98,v/v)at a flow rate of 1 ml/min and the detection wavelength was set at 405nm.RESULTS:The standard curve was linear over the concentration range of 50～1000nmol/ml for putrescine and spermine with correlation coefficient 0.9991 and 0.9992.The recoveries were 94.65% with RSD 1.6% and 90.48% with RSD 2.1%for putrescine and spermine,respectively.CONCLUTION:The present study provided a sensitive,accurate and reproducible method for the determination ofpolyamine levels in human saliva.更多还原
