β3肾上腺素能受体Trp/Arg突变与糖尿病视网膜病变的相关性研究

【摘要】 目的研究中国人β3肾上腺素能受体(β3AR)基因Trp64Arg突变与糖尿病视网膜病变(DR)的相关性。方法:随机选取住院2型糖尿病患者144例,其中男性60人,女性84人, BMI(25.2±3.9)kg/m2,病程均≥5年,根据眼底荧光造影结果分为增殖性糖尿病视网膜病变(PDR) 组、非增殖性糖尿病视网膜病变(NPDR)组及对照组(无糖尿病视网膜病变组)。用酶法测血脂, 用葡萄糖氧化酶法测血糖,放免法测血胰岛素,C肽,HPLC法测HbA1c。用PCR-RFLP法判断β3AR 基因型。结果:1)中国人β3AR基因突变率为18.6%,低于Pima印地安人的31%,而高于欧美等白色人种的7-12%,与日本人突变频率相似。2)与正常基因型相比,杂合子BMI(25.8±3.8vs25.0 ±3.4,P<0.05)增高,血糖、血脂、血胰岛素、C肽无明显差异。3)NPDR组、PDR组与对照组之间β3AR 64Arg等位基因频率无明显差异。结论:β3AR基因参与中国人体重的调节,β3AR Trp64Arg 突变与中国人DR发病无关。
【Abstract】 Objectives To investigate whether the polymorphism of the β3-adrenergic receptor (β3AR) gene was associated with diabetic retinopathy(DR) in Chinese type 2 diabetes patients. Methods 144 type 2 diabetes patients were devided into NPDR, PDR, control group according to fundus photographs results. The polymorphism of the β3AR was determined by polymerase chain reaction-restriction fragment length polymorphism analysis. Results 3 The gene frequency of Trp64Arg mutation in our subjects was 18. 6%. The Trp64Arg allele of the β3AR was not significant difference among the three groups. Those with the mutation had higher BMI values, compared with those without the mutation. Conclude Thepβ3AR gene polymorphism is associated with higher BMI but not associated with DR in Chinese type 2 DM.
